摘 要:目的··:研究三氟拉嗪对吗啡镇痛耐受性的影响。方法··:热板测痛法,测定三氟拉嗪对小鼠热板(55℃)痛阈值(s)的影响;慢性吗啡处理使小鼠对吗啡的镇痛作用产生耐受性 ,观察低剂量三氟拉嗪对吗啡耐受小鼠最大镇痛效率的影响。结果··:(1)三氟拉嗪(2-20mg·kg-1)呈剂量依赖性延长小鼠的热板(55℃)痛阈值(s) ;(2)慢性吗啡处理使6mg·kg-1 吗啡最大镇痛效率降低42.2%(P<0.05),9mg·kg-1 吗啡最大镇痛效率降低9.8%(P>0.05) ;(3)合用低剂量三氟拉嗪(2mg·kg-1)和吗啡(6mg.kg-1)可以使吗啡耐受小鼠的最大镇痛效率提高47.9 %(P<0.01),其最大镇痛效率与单独急性吗啡(6mg·kg-1)处理相同。Objective: To explore the effect of trifluoperazine on tolerance of morphine analgesia in mice. Method: With hot-plate test, the effect of trifluoperazine on pain threshold (s) in mice was investigated. Chronic treatment of morphine in mice yielded the tolerance to morphine analgesia and then the effect of low dose trifluoperazine on percentage of maximal possible effect(MPE%) in morphine-tolerated mice was observed. Result:(1) Trifluoperazine (2-20 mg·kg-1) dose-dependently increased pain threshold (s) in hot plate (55℃) test; (2) Chronic administration of morphine reduced 42.2% MPE in morphine 6 mg·kg-1 group (P<0.05);(3) Co-administration of trifluoperazine (2 mg·kg-1) and morphine (6 mg·kg-1) rose 47.9% MPE in morphine-tolerated mice, which was within the same range as the acute morphine 6 mg·kg-1 group. Conclusion: Trifluoperazine may reverse the tolerance of morphine analgesia in mice.
