摘 要:目的 探讨内源性表皮生长因子 (EGF)的变化规律及其与创面愈合的关系。方法 采用免疫组织化学、核酸分子杂交及图象分析等方法 ,分别在蛋白质和转录水平检测深Ⅱ度烫伤创面愈合过程中 ,小鼠体内EGF主要合成器官EGF及其mRNA的表达。结果 颌下腺EGF的表达量于伤后 1d较对照组明显降低 (P <0 .0 1) ,伤后 5d恢复至伤前水平并持续至伤后 2 1d ;其mRNA的表达强度伤前和伤后均为弱阳性~阳性。肾脏EGF的表达量于伤后 1d~ 5d较对照组明显降低 (P <0 .0 1) ,伤后 7d ,其表达量高于对照组并持续到伤后 2 1d(P <0 .0 1) ;其mRNA的表达强度伤前和伤后 3d均为弱阳性 ,伤后 7d~ 2 1d增强为阳性。伤后十二指肠EGF的表达量始终低于对照组 (P <0 .0 1) ;其mRNA的表达强度较对照组减弱。结论 在小鼠深Ⅱ度烫伤创面愈合过程中 ,颌下腺合成的EGF参与早期组织修复 ,肾脏合成的EGF参与创面的后期修复 ,十二指肠合成的EGF未参与皮肤烫伤创面的修复。Objective To investigate the change of endogenous epidermal growth factor and the relationship between this change and wound healing.Methods Immunohistochemisty, in situ hybridation and image analysis were employed to detect the expressions of EGF and its mRNA in organs mainly synthesized EGF in mice.Results The expression quantity of EGF in submaxillary gland was obviously lower than it in control group on the 1 st day post-scald ( P <0.01),and it would reach the pre-scald level on the 5 th day post scald and lasted till the 21 st day. The expression quantity of EGF in kidney was obviously lower than it in control group in the early five days post-scald ( P <0.01), and it would reach the pre scald level on the 7 th day post scald and lasted till the 21 st day ( P< 0.01). The mRNA expression was weak positive pre scald and the early 3 days post scald, and from the 7 th day, it became positive. In duodeum, the expression quantity of EGF was lower than it in control group during plst scald time ( P <0.01),and compared with control group, its mRNA expression was weakened.Conclusion During second deep degree scald wound healing in mice, EGF synthesized by submaxillary gland and kidney might participate in wound repair in the early and late stage respectively, and EGF synthesized by duodeum might not participate in wound repair.
