治喘贴对实验大鼠嗜酸粒细胞凋亡及对白介素-5的作用研究

摘　　要：目的 :研究治喘贴对卵清蛋白所致实验大鼠哮喘体内嗜酸粒细胞 (EOS)凋亡及对白介素 5 (IL 5 )的调节作用。方法 :将实验动物随机分为对照组、哮喘模型组、代温灸膏治疗组、治喘贴治疗组 ,经外贴治疗 18日。收集支气管肺泡灌洗液 (BAL F)并采集心脏血 ,采用流式细胞仪、免疫组织化学 (TU NEL)和酶联免疫吸附(EL ISA)法分析血中 EOS凋亡和 BAL F中 EOS凋亡数及 IL 5含量。结果 :哮喘大鼠血和 BAL F中 EOS凋亡数和 IL 5含量均高于对照组动物 (P均 <0 .0 1) ,治喘贴治疗大鼠 IL 5和 EOS均明显低于哮喘组 (P均 <0 .0 1)。结论 :治喘贴能促进哮喘大鼠体内 EOS凋亡 ,并降低 IL 5水平 。Objective:To investigate the effect of Zhichuantie(治喘贴) on eosinophils(EOS) and interleukin5 (IL5) of asthma in experimental animals(rattus norregicus) after being infected with ovalbumin.Methods:Experimental animals were randomly divided into four group,fitness group,asthma group,Daiwenjiugao(代温灸膏)treatment group,Zhichuantie treatment group.After 18 days of treatment,the bronchus alveoli lavage fluid(BALF) and heat blood of rattus norregicus were collected.Then we assayed the apoptosis of EOS in blood,the quantity of EOS and the level of IL5 in BALF through the flow cytometry(FCM),terminal deoxynuclectidyl transferasemediated dUTP nickendlabeling(TUNEL) and enzymelinked immunosorbent assay(ELASA).Results:The content of EOS, IL5 in blood and BALF in the asthma group were higher than which in fitness group (all P <0 01),Zhichuantie group was lower than any of group exceptment and fitness(all P <0 01).Conclusions:Zhichuantie has an apoptosis effect on EOS and it can reduce the level of EOS, IL5 .And it can improve airway allergy inflammation of asthma in rattus norregicus with ovalbumin.
