创伤小鼠注射内毒素后巨噬细胞Ⅰa抗原表达的变化

摘　　要：目的 :探讨内毒素注射对创伤小鼠巨噬细胞抗原提呈功能的影响。方法 :6 0只小鼠以双侧股骨骨折及软组织挤压致伤 ,伤后次日静脉注射内毒素 (1ml/ kg) ,对照组注射等量生理盐水。观察动物腹腔和肺泡巨噬细胞 a抗原的表达 ;测定血清白介素 6 (IL 6 )含量。结果 :创伤后小鼠巨噬细胞 a表达明显降低 ,伤后第 1日为最低水平 ,第 7日恢复至接近正常。注射内毒素后巨噬细胞 a抗原表达降低更为剧烈 ,持续在较低水平 ,伤后 7日仍显著低于伤前。伤后血清 IL 6含量显著升高 ,注射内毒素后升高更为显著。结论Objective:To investigate the effect of endotoxin on macrophage antigen presentation function in posttraumatic mice.Methods:Sixty Balb/c mice were injured by double femur fracture and soft tissue crash,endotoxin (1 mg/kg ) was injected intravenously on the first posttrauma day in the endotoxin group,while normal saline was injected in the control group.Macrophage Ⅰa expression was measured by flow cytometry and serum interleukin6(IL6) concentration was measured by enzymelinked immunosorbent assay(ELISA).Results:Macrophage Ⅰa antigen expression was markedly inhibited after trauma,it reached the lowest value on the first posttrauma day and returned gradually to normal range on the seventh posttrauma day.Endotoxin challenge further decreased the expression of Ⅰa antigens on macrophages,and it maintained a low level on the seventh posttrauma day.Serum IL6 concentration was obviously increased after trauma,it was much higher in the endotoxin group compared with controls.Conclusions:Endotoxin enhances the disturbance of macrophage antigen presentation function after trauma.
