摘　　要：目的 :探讨普托品 (protopine,PRO)的心血管药理作用 ,为研制治疗心血管疾病新药提供理论基础。方法 :采用放射性核素86Rb ,测定PRO对小鼠心肌营养性血流量的影响 ,同时研究了PRO对麻醉大鼠的降压作用 ,并观察PRO对小鼠耐常压缺氧时间和耗氧量的影响。结果 :PRO 2 0mg/kg和 40mg/kgip能明显增加小鼠心肌营养性血流量 ,而且与等剂量夏天无总碱 (corydalisamabilismigo ,COAMTA)的作用相似。PRO 0 .5、1及 2mg/kgiv的降压作用呈剂量依赖性 ,起效快、维持时间随剂量增大而延长。PRO 40mg/kgip能延长小鼠生存时间 ,并轻度降低耗氧量 ,作用与COAMTA无显著差异。结论 :PRO是COAMTA中一种具有明显药理活性的单体 。Objectives: To investigate the pharmacological effect of protopine (PRO) on cardiovascular system and provide theoretical basis for treating cardiovascular diseases. Methods: The effect of PRO on mice’s myocardial nutritional blood flow was detected by using 86 Rb. The hypotensive action of PRO was also observed in anesthetized rats. Moreover, the role of PRO on mice’s resistance to oxygen lack and the oxygen consumption was studied. Results: PRO, similar to COAMTA, 20mg/kg or 40mg/kg intraperitoneal injection could increase mice’s myocardial nutritional blood flow obviously. PRO at the dose of 0.5, 1 or 2mg/kg iv could lowered blood pressure significantly with dose-dependent and occurred quickly. PRO 40mg/kg intraperitoneal injection, similar to COAMTA, could prolong survival time of mice and slightly reduced oxygen consumption. Conclusions: PRO is one of the monomers that possess significant pharmacological activity in COAMTA, and has distinct effects in the development of new drugs to cardiovascular diseases.
