摘  要:目的 探讨T细胞亚群及共刺激分子表达与系统性红斑狼疮免疫病理的关系。方法 采用免疫荧光标记技术和流式细胞仪检测技术对系统性红斑狼疮 (SLE)患者和正常人外周血T细胞亚群及外周血单个核细胞膜表面共刺激分子的表达进行分析。结果 ①与正常人相比 ,SLE患者CD3+ 、CD4+ T细胞亚群显著减少 ,CD4/CD8比例失调 ,双标记CD4+ CD2 8+ 、CD8+ CD2 8+ T细胞亦显著减少 ;②患者共刺激分子CD2 8表达显著降低 ,4-1BB分子表达显著增高 ;③患者T细胞CD4/CD8与抗核抗体呈负相关 ,共刺激分子 4- 1BB与抗核抗体、尿蛋白呈正相关 ,而与CD4+ CD2 8+ T细胞亚群呈负相关。结论 T细胞亚群及共刺激分子表达异常在系统性红斑狼疮发病机制中可能具有重要作用。Objective To explore the relationship between T cell subsets, expression of costimulatory molecules and the immunopathy of systemic lupus erythematosus (SLE).Methods T cell subsets of peripheral blood and the expression of costimulatory molecules on the membrane surface of peripheral blood mononuclear cells in SLE patients and normal persons were detected by immunofluorescent lebeled technology and flow cytometry analysis.Results ①Compared with normal subjects, CD3 +,CD4 +,CD4 +CD28 +,CD8 +CD28 + T cell subsets were significantly decreased.The ratio of CD4/CD8 T cells was reversed; ②Significantly decresed expression of costimulatory molecule CD28 and increased expression of 4-1BB in SLE patients were observed; ③Negatived correlation was found between the ratio of CD4/CD8 T cells and the titer of ANA;On the contrary, the expression of costimulatory molecule 4-1BB was positively correlated with ANA and urine protein,while negatively correlated with CD4 +CD28 + T cell.Conclusion The changes of T cell subsets and abnormal expression of costimulatory molecules may have important affect on the pathogenesis of SLE.
