摘  要:目的 研究共刺激分子 (CD80和CD86)和主要组织相容性复合物 (MHC)在肝细胞癌 (HCC)细胞中的表达 ,探讨γ干扰素 (γ INF)治疗HCC中的免疫作用机理。方法 应用流式细胞仪测定CD80、CD86和MHCⅡ类分子HLA DR ,以及应用微量细胞毒方法测定MHCⅠ类分子在γ INF刺激前后 8种HCC细胞中的表达。结果 所有未受刺激的HCC细胞的CD80、CD86和HLA DR表达率均不超过 3 .5 8% ,只在 3种细胞中测到MHCⅠ类分子的表达 ;γ INF以 10 6U /L的浓度刺激 48h后 ,所有 8种HCC细胞HLA DR表达率均升高 ,其中 7种升高至 18%以上 ,5种升高至 70 %以上 ,荧光强度增强 10~ 5 0倍 ;刺激后 ,在 8种细胞中均可测到MHCⅠ类分子表达 ;刺激后 ,所有 8种HCC细胞CD86表达率均升高 ,7种升高至 12 .3 5 %~ 17.10 % ,5种HCC细胞CD80表达率升高至 5 %~ 10 %。结论 γ INF刺激可使多数被研究的HCC细胞获得一定的抗原呈递能力 ,同时可使所有被研究的HCC细胞获得被细胞毒T淋巴细胞识别和杀伤的条件。Objective To study the expression of costimulatory molecules (CD80 and CD86) and major histocompatibility complex (MHC molecules) by 8 hepatocellular carcinoma (HCC) cell lines under the stimulation of γ interferon (γ INF),to investigate the mechanism of γ INF in the treatment of HCC.Methods The expression of CD80,CD86 and MHC classⅡmolecule of HLA DR by HCC cells under the stimulation of γ INF at the concentration of 10 6?U/L for 48 h was detected by flow cytometry,and the expression of MHC classⅠmolecules by HCC cells detected by microcytotoxicity.Results The expression of CD80,CD86 and HLA DR by all 8 HCC cell lines was no more than 3.58%.The positive rate of HLA DR expression by BEL7404 and HpG2 cultured for 48 h in a medium supplemented with γ INF was increased up to nearly 20%,by BEL7405 up to 70.75%,by BEL7402,QGY7701,QGY7703 and SMMC7721 up to about 95%.The positive rate of CD86 expression by 7 HCC cell lines cultured with γ INF was increased to 12.35% to 17.10%.Without the stimulation of γ INF,MHC classⅠmolecules was positive in only 3 HCC cell lines,whereas under the stimulation of γ INF all 8 HCC cell lines expressed MHC classⅠmolecules.Conclusion γ INF significantly increased the expression of HLA DR and costimulatory molecules by most studied HCC cells and improved the ability of those HCC cells to present antigens;γ INF could promote the MHC classⅠmolecules expression by all studied HCC cells and the ability of HCC cells to be recognized by cytotoxic T lymphocytes.This might be the mechanism by which γ INF induces antitumor immunity in the treatment of HCC.
