绛红色小单孢菌噬菌体启动子在大肠杆菌中的克隆和表达

摘　　要：绛红色小单孢菌是氨基糖苷类庆大霉素、小诺霉素的产生菌。长期以来,人们对微生物的次级代谢作用进行了大量的研究,对小单孢菌也进行了细胞融合、质粒抽提及感染噬菌体等方面的研究。Sau3A restriction fragments from Microtyonospora purpurea phageMph1 DNA have been cloned into promoter probe plasmid pGA46 whichhad been digested with Bgl Ⅱ. Transforments were selected on mediumthat allowed the selection of T_c^r promoter bearing plasmid. The origin of the cloned promoter was confirmed by means ofSouthern blot hybridization. Analysis of the molecular structure of thepromoter got underway. We did not find that environmental factors suchas pH and NaCl concentration in medium had influence on expressionof promoter cloned from Micromonospora purpurea phage.
