成年猪胰岛的分离和纯化

摘　　要：目的 :观察胶原酶消化法对成年猪胰岛的分离效果。方法 :采用胶原酶胰导管注射负荷技术 ,静止消化与物理消化相结合消化分离成年猪胰腺脾叶 ;不连续密度梯度比重离心纯化 ,胰岛素释放试验检测胰岛细胞功能。结果 :胰岛收获量为 8339± 2 30 5胰岛数 /g胰腺 ,纯化后胰岛数 / g胰腺为 436 7± 1876 ,纯度为 85 %,胰岛素释放反应良好。结论 :采用本方法消化分离成年猪胰岛 ,胰岛细胞产量、纯度较高 ,功能良好。Objective:To observe the digestion effects of collagenase on adult porcine pancreas.Methods:Collagenase solution was injected through the pancreatic duct. The splenic lobe of porcine pancreas was digested by static digestion combined with physics digestion.Discontinuous density gradient centrifugation were used for purification of swine islet. The insulin secretion of islets was determined by Glucose-stimulated insulin release test.Results:The average was 8339±2305 islets per gram of pancreas. After purification the recovery was 4367±1876 islets per gram of pancreas.Purity of the final preparation was 85% islets. There was a good response to the glucose_stimulated insulin release test.Conclusions:The digestion of collagenase for adult pig pancreas has a high yield and high purity of islets which have biological vigor.
