摘 要:将染料分子偶联于交联琼脂糖疑胶上制成蓝色、红色和黄色等五种层析介质进行血清白蛋白分离的研究。比较了五种染料配基的吸附性能,考察了温度、pH值、蛋白质浓度和无机盐浓度对吸附血清白蛋白的影响。选择了合适的分离条件。从产妇分娩血中用染料配基吸附层析提取了人血清白蛋白(HSA)。一步分离达到电泳纯,回收率为95%,优于传统的盐析工艺。Dye molecules were coupled to agarose gel beads to form blue, redand yellow chromatography media for the separation of serum albumin.The adsorption properties of five dye-ligands were compared and theeffects of temperature, pH, protein concentration and salt concentrationupon the adsorption of serum albumin to the gels were investigated.Human serum albumin(HSA) was purified from human placental plasmaby one step of dye-ligand chromatography under suitable operatingcondition. The product was electrophoretically pure and the elutionrecovery was as high as 95%. This new technique is superior to theconventional process of salting-out.
