摘  要:目的 研究永生化髁突软骨细胞 (immortalizedmandibularcondylarchondrocyte,IMCC)的细胞周期及核型特征。方法 收集不同代数的IMCC和第 4代的髁突软骨细胞 (mandibularcondylarchondrocyte ,MCC)分别用流式细胞仪检测两种细胞的细胞周期特征 ,并用染色体显带法观察其核型特征。结果 G1期的MCC和IMCC所占百分比分别是 79.7%和 60 .0 % ,G2期的细胞分别为 6.1%和 2 1.2 % ,S期的细胞分别为 14 .3 %和 18.8%。第 4代MCC和第 60代、第 84代IMCC中有 4 4条染色体的细胞分别占 90 %、5 3 %和 3 1%。结论 IMCC细胞周期发生改变 ,细胞增殖速度加快 ;多数IMCC染色体维持在 4 4条 ,但随着培养次数的增加 。Objective To study the character of cell cycle and karyotype of immortalized mandibular condylar chondrocyte(IMCC).Methods Flow cytometry was used to detect the cell cycle of IMCC of different passage and mandibular condylar chondrocyte (MCC) of passage fourth.Trypsin-Giemsa staining method was also used to observe the karyotype of them.Results Flow cytometric analysis showed that G1 cells of 1MCC and MCC were 60.6 % and 79.7 %respectively,followed by 18.8 % and 14.3 % S cells,and 21.2 % and 6.1 % G2 cells in IMCC and MCC respectively.Karyotype assay found that 90% MCC of the fourth passage had 44 chromosome,but only 53% IMCC of 60 th passage and 31% of IMCC of 85 th passage had normal 44 chromosomes.Conclusion The cell cycle of IMCC has changed,and IMCC can proliferate more quickly than MCC.The majority of IMCC have the normal karyotype,which might be unstable following the passage.
