摘  要:用SPF鸡胚从我国东北地区猪群中分离出 3株H3 N2 猪流感病毒 (SIV) ,分别命名为A/Swine/Heilongjiang/ 1/ 2 0 0 0、A/Swine/Heilogjiang/ 2 / 2 0 0 0和A/Swine/Heilogjiang/ 3/ 2 0 0 0。SIV分离株第 8代病毒液对 0 .5 %豚鼠红血球的血凝价分别为2 0 48log2 、2 0 48log2 、40 96log2 ;将其制成琼扩 (AGP)抗原与禽流感标准阳性血清均出现清晰白色沉淀线 ;与流感病毒H3 亚型标准阳性血清的血凝抑制 (HI)价分别为 8log2 、7log2 、7log2 ,但不能被鸡新城疫阳性血清抑制。这 3株SIV都为热不稳定型 ;均能凝集驴、绵羊、鸡、兔、豚鼠、小鼠、大鼠、人O型血红血球 ,对马、牛红血球均不凝集 ;感染一个病毒最小致死量的鸡胚平均死亡时间分别为 86 .4、12 9.6和 91.2小时 ;每 0 .2ml病毒液鸡胚半数感染量分别为 4.75lg、4.5lg、和 6 .0lg ;对鸡的静脉内接种致病指数和脑内接种致病指数各为 0 .5 6、0 .32、0 .5 2和 0 .77、0 .38、0 .5 2 ;对小鼠的半数致死量分别为 2 .6lg、2 .0lg和 2 .5lg ;试验感染小鼠、豚鼠和猪均可发病并出现死亡。存活动物接种后第 6天和第 14天的血清 ,其HI价均在 8log2 以上 ,而AGP均为阴性。Three H 3N 2 subtype strains of swine influenza virus (SIV),which were referred to as A/Swine/Heilongjiang/1/2000 (Sw/HLJ/1),A/Swine/Heilongjiang/2/2000(Sw/HLJ/2) and A/Swine/Heilongjiang/3/2000(Sw/HLJ/3),are isolated from northeast of China.Under electron micrograph,virions reveal various shape of spherical with a diameter of 80~120nm as well as elliptic and elongated form.These isolates could grow and replicate in embryo,and the hemagglutinin(HA)values of harvested egg fluids(the 8 th generation)are 2048log2,2048log 2 and 4096log 2 respectively.Agar_gel precipitin(AGP)antigen make from SIV isolates could react with serum against avian influenza virus.The hemagglutinin inhibition(HI)titers of these strains against antiserum of H 3 subtype are 8log 2,7log 2 and 7log 2 respectively,whereas they are negative in HI assays against Newcastle disease antiserum.Three H 3N 2 viruses are not stability in 56℃water and shows HA activity to erythrocyteg of donkey,sheep,chick,rabbit,guinea pig,mouse,rat and O type blood of human,but only Sw/HLJ/3 strain could agglutinate the RBC of pig.Mean death time(MDT) of isolates are 86.4h,129.6h and 91.2h,while their 50% percent embryo infectious dose(EID 50) and 50% lethal dose(LD 50) are 4.75lg,4.5lg,6.0lg and 2.6lg,2.0lg,2.5lg,respectively.However,their intravenos pathogenicity index(IVPI) and intracerebral pathogenicity index(ICPI) are 0.56,0.32,0.52 and 0.77,0.38,0.52,respectively.Results above indicates that two isolates are moderate pathogenicity to chicken while the other one is lowly pathogenic,This shows that both of moderate and lowly pathogenic SIV exists in domestic pig farm.Furthermore,all of isolates could infect mouse,guinea pig and swine in lab.
