坏死梭杆菌毒力菌株FN(AB)94免疫原对鹿的初步免疫试验

摘　　要：本研究将坏死梭杆菌毒力菌株FN(AB) 94厌氧培养后 ,裂解制备抗原 ,与等量福氏完全佐剂混合 ,制备乳化佐剂疫苗。将疫苗分别接种 4头不同年龄的健康成年鹿 (每头接种 4ml)。 3头鹿作为对照组。接种前 ,静脉采血 ,进行对流免疫电泳检查。 30天后 ,分别用 15 0～ 40 0亿菌感染试验动物 ,然后混群饲养观察 ,并定期采血检查被免鹿血清中抗体消长变化。结果对照组鹿分别于 3个月后开始出现明显的临床症状 ,而免疫组不表现任何临床症状 ,6个月仍表现临床健康。 4头免疫鹿 6个月时都能检测到血清抗体 ,3头免疫鹿血清抗体可维持到 7个月后。试验表明 ,坏死梭杆菌毒力菌株FN(AB) 94疫苗接种鹿后 ,可刺激鹿产生很好的免疫力 ,从而通过本动物初步证明坏死梭杆菌毒力菌株FN(AB) 94免疫原具有良好的免疫原性 ,可以用于制备坏死杆菌病疫苗。Cultures of a virulent isolate Fusobacterium necrophorum biotype AB(FN(AB)94) from deer with necrobacillosis are sonicated,concentrated and mixed in an equal volume with the complete Freud′s adjuvant to develop an vaccine.A dose of the vaccine containing 4ml components made from 2×10 10cells of FN(AB)94 per ml culture grown in EY broth for 20 hours is respectively inoculated into 4 test deer,3 deer as controls.After 30 days,each test deer is inoculated with a different dose of 1.5 to 4×10 10cells of FN(AB)94 per ml fresh cultures of the homologous FN(AB)94 isolate.Each of 3 deer as controls is also inoculated with a different dose of 1.5 to 3.5×10 10cells of FN(AB)94 per ml fresh cultures of the homologous FN(AB)94 isolate.All test and control deer are raised in a same pen.Blood is collected from each test deer to examine the antibody level change.Results showed that all deer as controls occurred in a same symptom with striking swollen pustules appeared along the inoculated part of the neck,while test deer are all survived over 6 months without any abnormal signs.Twenty days after inoculation of the vaccine,antibody can be examined in blood of each test deer and lasted for 6 months.It can be derived from this result that the test deer inoculated with the vaccine made from the isolate FN(AB)94 acquired highly immunogenicity against the homologous live F.necrophorum AB.Thus,the sonicated FN(AB)94 vaccine is further confirmed to be efficacy against necrobacillosis in deer besides rabbits.
