摘  要:目的 阐明基质细胞在造血细胞体外扩增中的作用。方法 采用骨髓基质细胞培养、骨髓单个核细胞(BMMNC)体外液体培养和各种造血祖细胞集落培养技术,进行基质细胞支持条件下骨髓单个核细胞体外扩增的研究,并与单纯细胞因子支持下的细胞扩增进行比较。结果 在照射后基质细胞层存在的条件下,同时含有细胞因子组合的培养体系,对细胞总数和集落形成细胞数的扩增作用明显高于其它各组。结论 基质细胞对细胞因子作用下的造血细胞体外扩增有明显的促进作用。Objective To study the effect of bong marrow stromal cells on ex vivo hematopoietic progenitor expansion. Methods Bone marrow stromal cells culture, ex vivo bone marrow mononuclear cells(BMMNC) expansion, semisolid culture of colony forming unit-progenitor cells techniques were used to study the expansion of BMMNC. Results When cytokines were added to the systems of bone marrow mononuclear cells co-cultured with irradiated stromal cells,the number of total cells, CFU-GM and CFU-Cs were increased notably than those of the uncultured control group, stimulated only with cytokines group or simple co-cultured group. Conclusion The expansion system contained both co-cultured stromal cells and cytokins can mostly expand the total cells and progenitors, and the bone marrow stromal cells have promoting effect on ex vivo hematopoietic cells expansion.
