摘 要:目的探讨商业用腹膜透析液commercialperitonealdialysateCDS对腹腔巨噬细胞功能的影响。方法培养的小鼠腹腔巨噬细胞MΦ在含不同葡萄糖浓度15、25和42g/L的CDS中,经不同时间10、30和60min的暴露,观察MΦ还原MTT的能力及其NO的产量。结果低葡萄糖浓度(25g/L)暴露10min实验组,MΦ还原MTT的能力(A570nm值)为0.210±0.008,NO的产量为(9.1±1.3)μmol/L,均明显低于对照组(P<0.01)高葡萄糖浓度(42g/L)和较长时间(60min)暴露组,MΦ还原MTT的能力(A570nm值)为0.056±0.004,NO的产量为(5.7±1.1)μmol/L,与对照组相比较,改变最明显(P<0.01)。结论短时间CDS暴露,即可降低MΦ的活力及NO产生,这种作用与CDS中的葡萄糖浓度以及MΦ暴露于CDS的时间呈正相关。Aim to explore the effect of peritoneal dialysate(CDS)on functions of murine peritoneal macrophages. Methods Cultured peritoneal macrophages were exposed to peritoneal dialysate containing different concentration of glucose (15, 25 and 42 g/L) for different interval times (10 min, 30 min and 60 min). Then the nitric oxide (NO) produced by peritoneal macrophages,and the viability of peritoneal macrophages was tested was observed by MTT method. Results In low concentration of glucose (25 g/L) and 10 min exposing to peritoneal dialysate group, viability(A570nm=0.210±0.008)and nitric oxide production[(9.1±1.3)μmol/L] of macrophages were remarkably lower than those in control group(P<0.01). High concentration of glucose (42.5 g/L ) and 60 min exposing group viability (A570nm=0.056±0.004) and NO production [(5.7±1.1)μmol/L] had obviously changed as compared with the control group(P<0.01). Conclusion Short-time exposing to peritoneal dialysate may decrease viability and NO production of peritoneal macrophages, and these two effects are directly proportional to concentration of glucose in peritoneal dialysate and the interval they exposed to that.
