补肾活血和泻下及开窍活血方药对脑缺血再灌注肺损伤老龄大鼠ATP酶和自由基代谢的影响

摘　　要：目的从ATP酶活性变化和自由基损伤方面研究益元活血丹和大黄及血栓心脉宁对老龄大鼠脑缺血再灌注肺损伤防护作用的机制。方法老龄(20月龄以上)大鼠分为正常对照组、模型组、尼莫地平组、益元活血丹大小剂量组、大黄组、血栓心脉宁组,观察大鼠全脑缺血30min再灌注60min后肺组织病理和超氧化物歧化酶(SOD)、ATP酶活性和丙二醛(MDA)含量的变化。结果老龄模型组肺组织出现明显的病理损伤,益元活血丹大小剂量组和血栓心脉宁组及大黄组肺损伤均显著改善,其中以大黄组和益元活血丹大小剂量组效果更显著;与老龄对照组比较,老龄模型组血清MDA/SOD比值增高,肺Ca2+-ATP酶活性降低;与老龄模型组比较,益元活血丹大小剂量组、大黄组和血栓心脉宁组血清和肺组织中MDA/SOD比值降低,益元活血丹小剂量组肺Ca2+-ATP酶活性增高。结论脑缺血再灌注可导致老龄大鼠肺损伤,益元活血丹和大黄及血栓心脉宁对肺损伤保护机制与其拮抗自由基损伤有关,益元活血丹提高Ca2+-ATP酶活性明显。Objective:To study the protective effects and m echanisms of Yiyuanhuoxuedan(YHD),rhubarb and Xueshuanxinmaining(XXN)on the pulmonary lesion in the aged rats with cerebral ischemia reperfusion from the change of ATP -ase activity a nd free radical injury.Methods:The aged rats(20months or more)were divided into control group(ACG),model group(AMG),Nimodipine group,high and low dose s group of YHD(YHDH,YHDL),rhubarb group(RG)and XXN group.It was determined that the pathological change of the pulmonary tissue,the SOD and ATP -ase activity and the chan ge of the content of MDA in the rats with 60minutes reperfusion after 30minutes ischemia.Re-sults:The pathological change of the pulmo nary tissue was obvious in the AMG,but it was improved significant l in the YHDH,the YHDL,the XXN and the RG,especially in the RG,t he YHDH,the YHDL.Compared with the ACG.the seru m MDA /SOD ratio of the serum and pulmonary tissue decrease d in the YHDH,the YHD,the RG and the XXN compared with that in the ACG.Compared with the AMG,the Ca 2+-ATP -ase activity of pulmonary tissue increased in the YHDL.Conclusion:Cerebral iscemia reperfusion could lead to pul-monary lesion in the aged rats;the me chanism of the protective effect of YHD,RG and XXN on the pulmonary lesio n was correlated with the re-sisting free radical injury,and YHD could obviously increase the Ca2+-ATP -ase activity.
