摘　　要：目的 :建立新的原发性血小板减少性紫癜 (ITP)病人的血清学检测方法。方法 :采用免疫细胞化学法 ,用巨核细胞 血小板系Dami细胞株进行涂片 ,利用Dami细胞膜上的表面标志物GpⅡb/Ⅲa、GpⅠb、vWF等作为抗原 ,检测ITP病人血清中的血小板自身抗体。结果 :部分Dami细胞内可见红色沉淀颗粒 ,颗粒着色深浅不一 ,部分Dami细胞胞膜呈红色 ,ITP病人组红色沉淀颗粒明显多于正常人组和非ITP性血小板减少组。结论Objective: To establish and evaluate the method of serologic testing of patients with ITP.Methods:The immunocellchemical method of smear of megakaryocytic platelet cell line, Dami cell, was used and the membrane surface markers of Dami cell, such as G pⅡb/Ⅲa?G pⅠ b?vWF were regarded as antigens to detect the serum platelet autoantibodies of the patients. Results: The smears showed that part of cells membrane was red and there were many red precipitative granules in Dami cells with different stain intensity. It was obvious that the red precipitation granules in smears of patients with ITP were much more than those of normal persons and the patients with nonimmune thrombocytopenic purpura. Conclusion:The above results indicate that serum platelet autoantibodies of ITP can be detected specifically with the method.
