摘  要:目的 观察亚甲蓝光化学法对临床单袋人血浆中各种病毒的灭活能力 ,以及处理后对血浆正常成分的影响。方法 采用细胞感染法评价病毒的灭活效果 ,用生化、免疫测定法判定血浆成分的破坏情况。结果 在国产PVC血浆袋中加入 2 0 0 ml人血浆 ,然后加入光敏剂亚甲蓝使其终浓度为 1μm ol/ L时 ,再给以 2 0 0 0 0 1ux强度的红色光照射 ,作用 10 min后 ,可使 10 5.0 9TCID50 的 HIV、10 6 .2 5TCID50 的 VSV和 10 6 .50 TCID50 的 Sindbis病毒完全灭活 ;作用 30 min,血浆中主要成分的生化指标无显著变化 ,各种酶活性未发生明显改变 ,凝血因子 促凝活性 (F :C)仍大于 80 % ,血浆的抗原性无特殊变化。结论 该方法具有良好的灭活输血传播之病毒的能力 ,并能保证绝大部分血浆的正常活性。OBJECTIVE To observe the ability of inactivating viruses in clinical plastic bag human plasma and the influence of its normal function by methylene blue photochemical methods.METHODS Virus infected cell was used to test the virus inactivation effect and the biochemical and immunological techniques were used to test the activities of normal plasma ingredients.RESULTS Clinical single unit plasma(200ml) in China made PVC plastic bag can be inactivated by 1 μmol/L methylene blue with 20 000 1x of red light.After 10 min disinfected time, all the added indication virus at the titers of 10 5.09 TCID 50 of HIV,10 6.25 TCID 50 of VSV and 10 6.50 TCID 50 of Sindbis viruses could be inactivated absolutely. After 30 min inactivating time, all of the biochemical opponent indicators had no significant changes,and main enzyme activities were not inactivated,the Ⅷ∶C factor activity was still larger than 80%,the antigen of the total plasma protein had not changed significantly.CONCLUSIONS Methylene blue photochemical methods can be used to inactivate the main transfusion infected viruses, and the normal activities of plasma still keep at very high level.
