摘  要:目的 研究植物蜕皮激素 (Ecdystreone ,EDS)是否对脑损伤有保护作用 ,并探讨其可能的作用机制。方法 采用流体撞击致大鼠中度急性脑损伤模型 ;Wistar大鼠 5 0只随机分为假损伤组 ,损伤组 ,EDS的溶剂 (5 %乙醇溶液 )对照组 ,地塞米松(dexamethasone ,DXM)对照组 (地塞米松磷酸钠 5mg·kg-1) ,EDS治疗组 (EDS ,2 0mg·kg-1) ;以海马血流量、脑组织水含量、病理改变为指标观察蜕皮甾酮对急性脑损伤的疗效 ;从血循环内皮细胞 (CEC)、脑组织一氧化氮 (NO)及脂质过氧化 (MDA)方面探讨蜕皮甾酮的作用机制。结果 EDS组血流量、NO均较损伤组为高 ;EDS组的脑组织水含量、CEC和MDA则低于损伤组 ;EDS组皮层、海马的病理变化也较损伤组为轻 ,而与地塞米松对照组相似。结论 蜕皮甾酮可减轻急性脑损伤 ,产生保护作用 ;其机制在于减轻血管内皮损伤 ,以及增加脑一氧化氮、减少脑组织脂质过氧化 。OBJECTIVE: The present experiment aimed to study whether the ecdysterone derived from plant could protect against rat acute cerebral injury and to investigate its potential mechanism. METHODS: The experiments underwent in rat model of left moderate cortex fluid percussion brain injury. Fifty rats were randomly divided into sham-injury group, injury group, impregnant(5% alcohol) control group, dexamethasone (DXM, 5mg&middotkg-1) control group and ecdysterone (EDS, 20 mg&middotkg-1) group, respectively. The cerebral blood flow (CBF) in hippocampus and cerebral water content in frontal lobe, parietal lobe, and hippocampus were measured. The pathologic changes in cortex and hipppcampus were observed by light microscopy. The circulating endothelial cells (CEC) in blood, nitric oxide (NO) in brain and malondialdehyde (MDA) in brain were also detected. RESULTS: The values of CBF and NO were higher in EDS group than in injury group. The contents of cerebral water, CEC and MDA were lower in EDS group than in injury group. The pathological changes in cortex and hippocampus such as cerebral edema were less severe in EDS group than in injury group and similar in EDS group to DXM group. CONCLUSION: EDS alleviated the acute cerebral injury induced by fluid-percussion and produced protective effect. The mechanism of protective effect might be related to abating the injury of endothelium and anti-lipid peroxidation. It is necessary to further study the pharmacological effect of EDS.
