摘  要:建立从离体培养高效率再生植株的程序是用细胞遗传操作改良农作物的一个先决步骤。从栽培大再组织培养能再生植株的报道,近年日渐增多(可参考文献)。其中,获得再生植株效率比较高的报道。Soybean plants mainly from Northeast China grown in our phytotron under xenon-lamp 504 μE·m^(-2)·s^(-1) 12 hr daily illumination at 25±1℃ were used as experimental materials.(1) The regenerative response of isolated immature cotyledon of various soybean genotypes to MSN 10 and OR 1 media system was examined. The explants were cultured at 25±2℃ under a 12 hr photoperiod of white fluorescent light (3.5 kErg.cm^(-2). s^(-1)). Three thousand and five hundred re plicates of 23 cultivars and 7 advanced breeding lines were extensively tested. The reaults indicate that different genotypes varied obviously in plant regeneration effciency to the same culture method. (Table 1, and 2)(2) A reproducible method(1/2 MB procedure) for plants regeneration from cotyledon node has been developed. Pest regeneration was from reducing of addition of benzyladenine step by step when buds had been initiated. 1/2 MB procedure includes; (1) Culture explants on 1/2MS/5 BA medium for 3 weeks under a 12 hr photoperiod: (2) Cut into pieces(per 5—10 mm i. d.) and transfer to MS salts + B_5 organic components + BA 0.4 mg/1 medium for 1 month under light/dark cycle as above: (3) Cut into pieces, transfer into B_5+ BA 0.1mg/1+IBA 0.1mg/1 medium for 1 menth under a 12 hr photoperiod: (4) Transfer shoot 1—2 cm height to B_5 (14 hr photoperiod) for rooting, the plant regeneration efficiency (plantlet formed per explant) of 1/2 MB procedure was about 0.3 (Table 3). The regenerated plants were transplanted to pots and the R_1 generation of plants have set normal seeds.
