摘 要:目的 探讨枯否细胞在烧伤后对肝细胞的作用。方法 以 30 % 度烫伤大鼠为模型 ,采用枯否细胞与肝细胞联合培养体系 ,检测培养上清中 MDA、TNF、AL T、L DH的变化及添加 TNF单抗的影响。结果 烧伤后活化的枯否细胞与正常肝细胞及烧伤后受损肝细胞联合培养组上清中 AL T、L DH、MDA、TNF含量显著增高 ;在培养上清中加入 TNF- α单抗后培养上清中 TNF含量较对应组显著下降 ,但 AL T、L DH却无明显改变。结论 烧伤后肠源性内毒素等可激活枯否细胞 ,而活化后的枯否细胞在体外亦能持续合成并分泌毒性介质 (如氧自由基、TNF等 ) ,从而介导肝细胞的损害 ,且这种损伤作用对受损的肝细胞更明显 ;TNF对肝脏无直接损害效应 ,必须依赖于其他因素 ,如OBJECTIVE To investigate the potential role of Kupffer′s cells on hepatocytes following thermal injury in vitro. METHODS Rats were subjected to 30% total body surface area (TBSA) Ⅲ° burn injury. The Kupffer′s cells derived from burnt rats were co-cultured with hepatocytes, and the contents of MDA, TNF, ALT and LDH in culture supernatant were detected, and the effects of the addition of anti-TNF monoclonal antibody to culture growth medium were studied. RESULTS The contents of MDA, TNF, ALT and LDH in culture supernatant were obviously increased. Adding anti-TNF monoclonal antibody to culture growth medium, the level of TNF in culture supernatant was higher than that of the control group, meanwhile the levels of ALT and LDH were no changed. CONCLUSIONS It suggests that Kupffer′s cells derived from burnt rats directly damage the hepatocytes in vitro, the damage is associated with the inflammatory cytokines (for example, reactive oxygen species, TNF) released by activated Kupffer′s cells. The toxic effect of TNF on hepatocytes is indirect, rather via synergistic action with other factors, such as PMNs.
