小鼠联会复合体超微结构畸变的观察与分析

摘  要:目的 :用改进的微铺展技术观察分析小鼠联会复合体超微结构的畸变。材料和方法 :以滨州医学院实验动物室饲养的性成熟雄性小鼠为材料 ,制备小鼠精母细胞联会复合体样品 ,硝酸银染色 ,电镜观察、分析。结果 :有联会复合体结构畸变和配对异常的细胞占观察细胞总数的 1 2 4%。其中联会复合体断裂占异常联会复合体总数的5 2 1 7% ,侧生组分断裂 3 1 5 2 % ,易位 6 5 2 % ,联会紊乱 4 3 5 % ,不联会 3 2 6% ,圆环 2 1 7%。结论 :实验小鼠各类联会复合体畸变百分率均高于国内文献报道的正常小鼠的联会复合体畸变率。改进的微铺展制备联会复合体样品技术简便易行 。Objective:To observe and analyze synaptonemal complex (SC) ultrastructural aberration of mice with an improved micro spreading technique.Methods:Mature male mice bred in the animal laboratory of Binzhou Medical College were used in the experiment.Synaptonemal complex in mice spermatocytes was prepared and silver stained,then was examined and analyzed by electron microscopy.Results:The cells with structrural aberrations and pairing abnormalities of SCs were 12.4% of observed cells,Breakages were 52.17% in the total abnormal SCs,lebreakages 31.52%,translocations 6.52%,synapsis irregularities 4.35%.Conclution:Asynapses of all kinds of SCs aberrationa are higher than normal mice reported in Chinese.The experiment indicates that the improved microspreading technique for the preparation of SCs samples is easy,and the result is good.
