摘  要:凝血酶适配分子 (aptamer)可与凝血酶以高亲和力结合 ,二者的结合可改变荧光素标记的适配分子溶液的荧光偏振强度 ,据此建立了适配分子荧光偏振法测定凝血酶的新方法。方法的线性范围是 0 .1~ 4IU/mL ;检出限为 0 .0 9IU/mL。将该方法用于人体血浆中凝血酶含量分析 。The anti-thrombin aptamer can interact with thrombin in high affinity. This interaction results in the change of the fluorescent polarization in the solution of the aptamer labeled with fluorescence. We developed a new method based on the change of the fluorescent polarization resulted from this interaction to detect thrombin. The linear range of the determination is 0.1 similar to 4 IU/mL. The detection limit for thrombin is 0.09 IU/mL. This method has been used to determine thrombin in human whole blood. The results obtained are satisfactory.
