褐飞虱单克隆抗体的制备及其初步应用

摘　　要：应用杂交瘤技术,制备了4株褐飞虱的单克隆抗体,分别命名为3B2、3B11、3H3和4B8。其只与褐飞虱所有虫态发生反应,不与稻田其它主要昆虫和蜘蛛发生交叉反应,具有高度的特异性。应用4B8初步检测浙江大学华家池校区农场稻田蜘蛛对褐飞虱的阳性反应率,其中拟环纹豹蛛、拟水狼蛛和纵条蝇狮的阳性反应率最高,粽管巢蛛、黑肩绿盲蝽次之,狡蛛较低,而八斑鞘腹蛛、跳蛛、肖蛸和食虫瘤胸蛛未检测到阳性反应。拟环纹豹蛛对4B8的阳性反应率与褐飞虱和拟环纹豹蛛种群密度均极显著相关（r=0.612＊＊,r=0.632＊＊）。4 monoclonal antibodies (McAbs) were developed to the whole homogenates of the brown planthopper, Nilaparvata lugens(St(?)l) using hybridoma technology.These McAbs,named 3B2,3B11,3H3,4B8, were highly specific to all the stages of N.lugens. They did not cross-react with any other insect species tested with an indirect enzyme-linked immunosorbent assay (ELISA).Using 4B8,the predator species from Hangzhou single cropping rice fields were tested.The positive rates of P irata subpiraticus, P ardosa pseudoannulate and M irpissa magister were the highest,those of C lubiona japonicola and Cyrtorrhinus were higher but those of C oleosoma octomaculatum,Salticidae, T etragnatha spp .and U.mmelita insecticeps were no positive reaction.Correlation analysis indicated that the positive reaction rate of P irata pseudoannulate to 4B8 had a significant relation to the poulatron of N.lugens and P. pseudoannulate (r=0.612**,r =0.632**).
