摘  要:本文研究人类轮状病毒基因DNA免疫及应用。通过构建重组质粒pCI/vp7,pCI/vp4及pCI/vp6,以肌注法导入BALB/c小鼠肌肉组织,其中pCI/vp7及pCI/vp4能 有效引起机体免疫应答,而且对轮状病毒Wa株有一定中和效应。从本次试验的结果来看 ,人类轮状病毒DNA疫苗能作为预防病毒性小儿腹泻的一种手段。The probability of DNA vaccine against human rotavirus wa s studied, by constructing three recombinant plasmid clones, pCI/vp7, pCI/vp4, a nd pCI/vp6, and transferring into TA muscle(quadriceps) of BALB/C mouse with intr amuscular inoculation. The sera were collected and antibody activity was evaluat ed with ELISA and neutralizing test. pCI/vp7, pCI/vp4, and pCI/vp6 have been suc cessfully Constructed, moreover, pCI/vp7 has transfected 293 cells and the vp7 gene could express thoroughly. Intramuscular inoculation of these plasmids induced ef ficient immuniation, and the antibody induced by pCI/vp7 presents some definite protective of a strain of rotavirus Wa. As the results showed, DNA vaccine will b e a helpful means to protect children from severe acute diarrhea caused by Group A rotaviruses.
