摘 要:利用大鼠离体肝脏灌流技术以及大鼠原代培养的肝细胞研究了大豆黄酮对SD大鼠肝脏氮代谢和IGF -I生成的影响。结果表明大豆黄酮可使大鼠离体肝脏灌流液中尿素氮浓度下降 ,并有一定的剂量效应和时间效应。在给药后灌流的第 1、2、3、4h ,1× 10 -4mol/L剂量的大豆黄酮试验组 (n =8)大鼠离体肝脏灌流液中尿素氮水平分别为 (0 .76± 0 .19)、(1.16± 0 .0 7)、(1.2 8± 0 .13)、(1.5 9± 0 .19) μg/ml(以含每毫克DNA的肝组织所生成的尿素氮浓度表示 ) ;与对照组相比 ,其尿素氮浓度下降的幅度分别为 13.6 4 % (P <0 .0 5 ) ,2 3.18%(P <0 .0 5 ) ,34.0 2 % (P <0 .0 1)和 32 .91% (P <0 .0 1)。 4× 10 -5mol/L的大豆黄酮对大鼠肝脏尿素氮生成影响具有类似的效应。大豆黄酮还可在一定程度上抑制大鼠离体灌流的肝组织中GPT的活性 (n =8) ,能够促进大鼠肝细胞蛋白质的合成 ,1× 10 -5mol/L的大豆黄酮可使3 H -亮氨酸的参入量与对照组相比提高了 2 1.38%(n =6 ,P <0 .0 5 )。大豆黄酮对离体灌流的大鼠肝脏IGF -I生成和分泌也具有一定程度的促进作用 (n =8)。提示大豆黄酮可通过增加对大鼠肝脏氮的储留、促进大鼠肝细胞蛋白质的合成以及在一定程度上影响肝脏IGF-I生成和GPT的活性 。The techniques of isolating perfused rat liver in vitro and culturing rat hepatocytes were used to study the effects of daidzein on liver nitrogen metabolism and IGF I level of Sprague Dawley rat. The results showed that urea nitrogen concentration of perfused medium was significantly affected by daidzein in a dose dependent and time dependent manner. The urea nitrogen concentration was 0.76± 0.19, 1.16±0.07, 1.28±0.13 and 1.59±0.19 μg/ml( n =8) for 1 h, 2 h, 3 h, 4 h after administering daidzein at the dose of 1×10 -4 mol/L, respectively; and the extent of decrease was 13.64% ( P <0.05 ),23.18% ( P <0.05),34.02% ( P <0.01) and 32.91% ( P <0.01), respectively, comparing with that of the control. The daidzein at the dose of 4×10 -5 mol/L had the similar effect on urea nitrogen level. GTP activity of isolated perfused rat liver was inhibited by daidzein, but the difference was not significant compared with control ( n =8). The protein synthesis of rat hepatocyte was enhanced by daidzein, the content of 3 H leucine incorporation into rat hepatocytewas significantly increased by 21.38% ( n =6, P < 0.05) with daidzein treated (1×10 -5 mol/L). IGF I production and secretion of rat hepatocyte were also affected by daidzein, but no significant difference was found compared to control ( n = 8). It suggested that daidzein could regulate nitrogen metabolism of rat liver by means of increasing nitrogen retention, enhancing synthesis of protein and affecting IGF I production and GTP activity of rat liver.
