摘  要:研究了苦槠种子和离体胚在超低温保存过程中含水量、冷冻方式、解冻方式 ,以及离体胚冰冻保护剂对电导率的影响 .结果表明 :含水量是超低温保存成功的关键 ,适度脱水可大幅度降低苦槠种子和离体胚超低温保存过程中的质膜伤害 ;随着含水量的降低 ,其电导率的变化呈先降后升 ,15 %含水量的电解质外渗率最低 ;而在冰冻保护剂作用下 ,离体胚保存最佳含水量为 10 % ,其质膜所受的伤害比较轻 ,电解质外渗量较小 。Effects of moisture content(MC), freezing and thawing types and cryopretectant pretreatment on the conductivity of Castanopsis sclerophylla seeds and excised embryos during cryopreservation were studied. The results were as follows:MC was the key factor of a successful cryopreservation, as the moderate dehydration could reduce the membranous injury of cryopreserved materials largely; with the MC declination, the membranous conductivity declined firstly, and then increased, the electrolyte leakage was the smallest under the condition of 15% MC without cryoprotectant; in contrast, by cryopreservation the optimal storage MC of excised embryos was 10%, as both the membrance injury and the leaked electrolyte were less, and the ability to amend membrane was higher than any other treatments.
