摘 要:利用放射自显影技术检测了小鼠着床前胚胎 ( 4-细胞、8-细胞、桑葚胚和囊胚 )不同发育阶段 (早、中、晚 ) S期和 M期卵裂球的比率 ,并研究了低温 ( 4℃ )对小鼠胚胎发育及 DNA合成的影响。结果显示 ,早期和中期 4 -细胞胚胎 S期卵裂球比率较高 ,分别是 75%、92 % ,没有 M期卵裂球 ,晚期 4 -细胞胚胎 S期卵裂球比率较低 ( 30 % ) ,M期卵裂球比率为 1 1 % ;早期和中期 8-细胞胚胎 S期卵裂球比率仍很高 ,分别是 73%、80 % ,M期卵裂球分别为 0、2 .5% ,晚期 8-细胞胚胎 S期卵裂球比率增至 6 4% ,M期卵裂球减少至 9% ;早、中、晚期桑葚胚的 S期卵裂球比率分别是 77%、72 %和 79% ,M期卵裂球的比率分别是 5%、3%和 3% ;早、中、晚期囊胚的 S期卵裂球比率分别是 78%、74 %和 77% ,M期卵裂球的比率分别是 4 %、3%和 4 %。 4℃的低温对小鼠早期胚胎的发育和 DNA合成都没有明显影响 ;4 -细胞、8-细胞胚胎和桑葚胚经低温处理及培养后其囊胚形成率和平均卵裂球数分别是 6 2 %和 ( 2 6± 6 )个、54%和 ( 2 5± 7)个、87%和 ( 34± 8)个 ;4 -细胞、8-细胞胚胎、桑葚胚和囊胚经低温处理后 S期卵裂球比率分别是 97%、77%、70 %和 74 %。The ratio of S phase and M phase of 4 cell,8 cell,morula and blastocyst in early,middle and late stage was determined by embryo autoradiographic technique,and the effect of low temperature on development and DNA synthesis of mouse embryos was studied.The results showed that the ratio of S phase of 4 cell embryos in early,middle and late stage is 75%,92% and 30% respectively and the ratio of M phase is 0,0 and 11% respectively.The ratio of S phase of 8 cell embryos in early,middle and late stage is 73%,80% and 64% respectively,and the ratio of M phase is 0,2 5% and 9% respectively.The ratio of S phase of morula in early,middle and late stage is 77%,72% and 79% respectively,and the ratio of M phase is 5%,3% and 3% respectively.The ratio of S phase of blastocyst in early,middle and late stage is 78%,74% and 77% respectively,and the ratio of M phase is 4%,3% and 4% respectively.Low temperature(4℃) has no effect on the development and DNA synthesis of mouse embryos.After the 4 cell embryo,8 cell embryo and morula are treated with low temperature and cultured in vitro ,the ratio of blastocyst formed and the average number of blastomeres is 62%(26±6),54%(25±7),87%(34±8) respectively.The ratio of DNA synthesis is 97%,77%,70% and 74% respectively.
