摘 要:目的 研究高温对小鼠胸腺细胞亚群及其热休克蛋白 70 (HSP70 )表达的影响 ,探讨热锻炼诱导小鼠胸腺热耐受形成的机制。方法 BALB/c小鼠每天 41℃高温处理 1h ,连续 9d ,于第 1、3、5、7、9天高温后 4h取胸腺细胞 ,用流式细胞术检测胸腺细胞亚群及HSP70表达阳性率。结果 高温使小鼠胸腺重量明显减轻 ,第 1、3天胸腺细胞CD4+CD8+亚群明显减少 ,第 5天后CD4+CD8+亚群比例开始恢复。除第 9天外 ,高温组胸腺细胞HSP70的表达均高于对照组。结论 CD4+CD8+胸腺细胞亚群减少可能是高温致小鼠胸腺萎缩的主要原因 ,而HSP70表达升高是热耐受形成的分子基础之一 。Objective To study the relationship among the change of thymocytes subsets, the expression of HSP70 and the thermotolerance mechanism of thymocytes during hyperthermia. Methods The mice were exposed to hyperthermia with hot air (41℃, relative humidity 70%) in a thermal chamber for 1h each day, and were killed 4h after heat stress on 1,3,5,7,9 day. After being weighed, thymuses were dispersed as single cells. The positive rates of thymocytes subsets and the HSP70 were assayed by flow cytometry. Results The weight of thymus was shown to be decreased during hyperthermia. the CD4 +CD8 + thymocyte subsets decreased at 1d and 3d, but increased since 5d. The positive rates of HSP70 in thymocytes from hyperthermia treated mice were higher than those of the controls except 9 days. Conclusion The decrease of thymus weight might possibly be due to decrease of CD4 +CD8 + thymocytes caused by hyperthermia. Elevation on HSP70 expression might be the molecular basis of thermotolerance formation and it might serve as a biomarker of thymocytes injury.
