摘 要:目的 应用分子流行病学手段探索男性不育的病因 ,为开展无精少精不育患者单精子注射技术的种植前遗传学诊断与筛选提供实验依据。方法 用质粒克隆技术对 RBM 1基因进行克隆、测序及 PCR技术检测 RBM 1基因的微小缺失。结果 在 75例男性不育患者中检测出 5例患者存在 RBM 1基因的微小缺失 ,缺失率为 6 .7% ,其中 10例无精症病例有 1例缺失 ,6 5例少精症患者中有 4例缺失。结论 我国正常男子人群中存在 RBM 1基因 。Objective To study etiological factors for male infertility by using molecular epidemiologic method and to provide experimental basis for genetically diagnosing and screening in azoospermic and oligozoospermic patients before they were carried out with Intracytoplasma Sperm Injection. Methods RBM1 gene was cloned by using plasmid cloning technique, sequenced and detected by PCR. Results Five patients were found RBM1 gene microdeletions among 75 male infertility patients, one in 10 azoospermia and four in 65 oligozospermia. The deletion rate was 6.7% . Conclusions RBM1 gene exists in Chinese males and there are RBM1 gene microdeletions in cases with azoospermia or oligozoospermia.
