摘 要:目的 测定大鼠内毒素血症期间淋巴细胞释放和合成降钙素基因相关肽 (CGRP)的变化。方法 采用逆转录聚合酶链反应 (RT- PCR)测定大鼠淋巴细胞 CGRP m RNA水平。用放射性标记法测定血浆 CGRP水平。结果 注射内毒素大鼠血浆和淋巴细胞释放 CGRP均增高 ,淋巴细胞中 CGRP m RNA表达也明显增高。结论 提示内毒素不仅刺激淋巴细胞释放 CGRP,而且能通过某些机制激活淋巴细胞 CGRP m RNA的转录 ,使 CGRP合成增加 ,以作为 CGRP大量释放的重要补充来源。Objective To investigate the changes of both CGRP release and synthesis during endotoxemia.Method Expression of CGRP mRNA in lymphocytes was determined by the semi quantity RT PCR and the CGRP levels in plasma were measured by RIA.Results Signficantly increased CGRP release in plasma and CGRP expression in lymphocyte were foundr 3hr and 8hr after endotoxin(5mg/kg,i.v.) was administrated.Conclusion It suggested that endotoxin could not only stimulate the release of CGRP from lymphocytes,but also the transcription of CGRP mRNA and synthesis of CGRP in lymphocytes during the development of endotoxemia in rats,so lymphocytes might be an important source of CGRP release during endotoxemia.
