摘  要:目的 建立一种新的丙酮酸氧化酶偶联比色法测定血浆丙酮酸。方法 利用丙酮酸氧化酶氧化丙酮酸生成过氧化氢 ,过氧化氢与 2 ,4 ,6-三溴 - 3 -羟基苯甲酸和氨基安替比林在过氧化物酶的作用下生成红色化合物。结果 该法线性范围达 10 0 0 μmol/L ,批内和批间平均变异系数分别为 1 77%和 2 2 3 % ,平均回收率 99 9% ,与紫外酶偶联法比较 ,相关系数r =0 .9986,y=1.0 0 3x + 0 .85 4 ,4 0例健康人血浆丙酮酸 3 2~ 10 4 μmol/L( x± 2S) ,15例糖尿病患者的血浆丙酮酸为 88 4 5± 3 1 98μmol/L。 结论 该方法具有灵敏、准确、简便、快速等优点。糖尿病患者血浆丙酮酸水平显著高于正常对照组 (P <0 0 1)。Objective To develop a coupling colorimetric method for assay of plasma pyruvate with pyyruvate oxidase.Methods Pyruvate oxidase was used as indicator enzyme,then the generated hydrogen peroxide was measured by use of peroxides with 2,4,6 tribromo 3 hydroxybenzoic acid and 4 aminoantipyrine.Results The standard curve for the assay was linear to 2 mmol/L.The average between run and within run coelticients of variation were 1.77% and 2.23%,respectively.The average recovery was 99.9%.Compared with ultraviolet method,r=0.9986,y=1.003 x+0.854,n=43.The values of pyruvate in 40 healthy subjects were 31~104 μmol/L(±2 s),and those in 15 diabetic patients were 88.45±31.98 μmol/L.Conclusions The method is sensitive,accurate,simple,rapid and suitable for clinical use.The level of pyruvate is significantly increased in diabetic patients (P<0.01).
