石榴皮抗Ⅱ型生殖器疱疹病毒的体内外实验研究

摘　　要：用体外细胞培养和实验动物感染模型观察了石榴皮抗Ⅱ型生殖器疱疹病毒（ＨＳＶ－２）作用。体外石榴皮提取液具有多环节的抗ＨＳＶ－２活性，尤以直接灭活和阻碍ＨＳＶ－２吸附效果明显。石榴皮栓剂经阴道内预防性用药，能防止ＨＳＶ－２感染的发生，小剂量石榴皮栓剂（５０ｍｇ）可减轻临床损害，阴道内病毒分泌减少，血清抗体滴度降低。大剂量石榴皮栓剂（１００ｍｇ）则能完全抑制ＨＳＶ－２感染，且ＰＣＲ检测未发现潜伏ＨＳＶ－２ＤＮＡ存在。 Antiviral effects of Punica granatum L.(PGL) against HSV-2 infection were observed with cell culture and animal model. The results of plaque reduce assay showed that an aqueous extract from PGL have multiple-effective targets at HSV-2 infection, especially in direct inactivity and blocking adsorption to cell. The results of animal infective model suggested that PGL vaginal suppository could effectively control the happen of HSV-2 infection by intravaginal preatreatment. The small dose group of PGL (50mg) partly inhibited HSV-2 infection. Compared with virus control group, the clinic lesions was alleviated and excretion of virus from vaginal tract was reduced and serum HSV-2 IgG antibody titer was decreased in guinea pigs.The large dose group of PGL(100mg) completely inhibited HSV-2 infection and latent HSV-2 DNA were not detected in the nervous tissues with PCR technique.
