API_(0134)对凝血酶诱导内皮细胞表达c-sis mRNA/PDGF-B链蛋白的影响

摘　　要：用免疫组化和原位杂交方法观察了ＡＰＩ０１３４（ＡＰＩ）对凝血酶诱导猪主动脉内皮细胞表达原癌基因ｃｓｉｓｍＲＮＡ和血小板源生长因子（ＰＤＧＦ）Ｂ链蛋白的影响。结果表明，凝血酶能诱导内皮细胞ｃｓｉｓｍＲＮＡ转录和促进ＰＤＧＦＢ链蛋白表达，ＡＰＩ则能显著抑制凝血酶的上述诱导作用，剂量增大抑制作用增强。 Objective:The effects of API0134 on expression of csis mRNA and plateletderived growth factor B chain protein(PDGFB) in porcine aortic endothelial cells (EC) induced by thrombin were studied in this article.Methods:Immunohistochemistry,in situ hybridization methods were used.Results:The thrombin induces csis mRNA transcription and stimilates PDGFB expression in EC;The inducible effects of thrombin can be inhibited by API0134 significantly,larger the dose,stronger the inhibitive effect.Conclusion:These data suggest that API0134 may play a role in antiatherosclerosis and prevention of restenosis after angioplasty.
