摘 要:目的:比较研究补阳还五汤和其有效部位组方抗脑缺血后DND 作用与脑组织能量代谢、NO 和NOS 的关系。方法:采用沙鼠双侧颈总动脉夹闭脑缺血再灌注模型,于再灌注后48 小时取前脑皮质测定Na +K+ATP 酶、Ca2 +ATP 酶、Mg2 +ATP 酶、乳酸、NO 和NOS。结果:缺血再灌注后48 小时脑组织Na+K+ATP酶、Ca2 +ATP 酶及Mg2 +ATP 酶活性均降低,NO 含量和NOS 活性降低,而乳酸含量无明显变化。补阳还五汤和有效部位组方ip 均可防止缺血再灌注48 小时后Na+K+ATP 酶活性的降低,增加NO 含量,提高NOS 活性,而有效部位组方还可增加缺血再灌注后Ca2 +ATP 酶和 Mg2 +ATP 酶活性,且其升高NO 的作用强于补阳还五汤组。结论:脑缺血后迟发性神经元坏死与脑内能量代谢障碍和NO 的生成减少有一定的关系,补阳还五汤及其有效部位组方可能通过调节脑内能量代谢和NOObjective: Effect of Bu Yang Huan Wu Tang (BYHWT) and its available composition combination antagonisming delayed neuronal death (DND) induced by cerebral ischemia and relation between DND and cerebral tissue energy metabolism, nitric oxide (NO) and nitric oxide synthase (NOS) will be studied. Method: cerebral ischemia model was made by ligating bilateral arteries carotis communis for 20 min followed reperfusion 48h in gerbils. Na + K + ATPase activity, lactic acid content, Ca 2+ ATPase, Mg 2+ ATPase activity, NO content and NOS activity were determined in fore brain cortex at 48h after reperfusion. Result: Na + K + ATPase, Ca 2+ ATPase and Mg 2+ ATPase activity lowered (P<0.01), NO level decreased (P<0.01), NOS activity dropped (P<0.01) in cerebral tissue at 48h after reperfusion, but lactic acid level had no change. BYHWT could prevent reduction of Na + K + ATPase activity (P<0.05, P<0.01), increase NO level (P<0.01), enhance NOS activity (P<0.01) at 48h after reperfusion. Conclution: DND induced by ischemia may be concerned with energy metabolism disorder and decrease of NO formation, BYHWT may play the role of antagonisming cerebral ischemia by adjusting cerebral energy metabolism and nitric oxide formation.
