摘 要:利用离体肝脏灌流技术测定了 CL(二个实验组 ,CL给药量分别为 1× 1 0 - 8mol和 1× 1 0 - 6mol)对大鼠肝脏物质代谢及相关酶活性的影响 .结果表明 CL可使大鼠离体肝脏灌流液中尿素氮浓度下降 ,并有一定的剂量效应和时间效应 .在给药后灌流的 1、2、3、4 h内 ,与对照组相比 ,1× 1 0 - 6mol的 CL使大鼠离体灌流的肝脏产生的尿素氮分别下降了1 4 .79% ( P>0 .0 5 ) ,1 7.88 ( P>0 .0 5 ) ,2 6 .79 ( P<0 .0 5 )和 2 9.95 ( P<0 .0 1 ) . 1× 1 0 - 8mol的 CL具有类似的效应 .而 CL对大鼠离体肝脏灌流液中葡萄糖的水平影响不大 .CL可抑制大鼠肝组织中 GPT的活性 ,灌流 4 h后 1× 1 0 - 6mol的 CL使得肝组织中 GPT活性下降了 2 4 .6 5 % ( P<0 .0 5 ) ,但 1× 1 0 - 8mol的 CL仅使 GPT活性下降 7.5 0 % ( P>0 .0 5 ) .CL 还抑制大鼠肝组织中 G6 PDH的活性 ,1× 1 0 - 6mol的 CL可使肝脏中 G6 PDH活性下降 36 .0 4 % ( P<0 .0 5 ) ,同样1× 1 0 - 8mol的 CL对 G6 PDH活性影响不显著 .提示 CL可直接通过影响肝脏的氮代谢及相关酶的活性继而影响机体的物质代谢 .The technique of isolated perfused rat liver in vitro was used to study the effects of clenasma (two treatment groups, the dose of clenasma at 1×10 -8 mol and 1×10 -6 mol , respectively)on metabolism and related enzyme activity in rat liver. The results showed that urea-nitrogen concentration of perfused medium was significantly affected by clenasma in a dose-dependent and time-dependent manner. The urea-nitrogen level was decreased by 14.79%( P>0.05),17.88%(P>0.05),26.79%(P<0.05) and 29.95%( P <0.01) for 1 h, 2 h, 3 h, 4 h after administering clenasma at the dose of 1×10 -6 mol, respectively, compared to that of control. 1×10 -8 mol clenasma had the similar effect on urea-nitrogen level. But glucose concentration of perfused medium was not affected by clenasma. GTP activity of isolated perfused rat liver was inhibited by clenasma. The enzyme activity was decreased by 24.65%( P <0.05) in 1×10 -6 mol clenasma-treated 4 h, however, GTP activity was only decreased by 7.50%( P >0.05) with clenasma-treated at the dose of 1×10 -8 mol. G6PDH activity of isolated perfused rat liver was also decreased by 36.04%( P <0.05) in 1×10 -6 mol clenasma-treated 4 h, and the enzyme activity of isolated perfused rat liver was not notably affected by the dose of 1×10 -8 mol clenasma. It is suggested that clenasma may improve metabolism by means of affecting nitrogen metabolism of liver and relation enzyme activity.
