摘 要:目的对比不同运动方式对自发性高血压大鼠(SHR)心脏重塑和运动能力的影响。方法30只雄性SHR随机分为安静组(SHR-s)、间歇运动组(SHR-IT)和持续运动组(SHR-CT),以10只Wistar-Kyoto(WKY)大鼠作为正常血压对照。实验后,测定大鼠运动能力、尾动脉血压、心脏结构与功能,HE染色和Masson染色进行组织病理学观察,PCR检测心肌相关基因mRNA表达。结果与WKY组比较,SHR-S组血压升高、心脏肥厚及胶原过度沉积,胚胎基因表达上调、收缩蛋白表达下调,心功能和运动能力下降(P<0.05);与SHR-S组比较,SHR-IT组和SHR-CT组血压下降、心脏肥大与心肌纤维化减轻,胚胎基因表达下调、收缩蛋白表达上调,心功能和运动能力改善(P<0.05);组间比较,SHR-IT组运动能力高于SHR-CT组(P<0.05)。结论(1)间歇运动和持续运动均可降低SHR安静血压水平、抑制心脏重塑并提高心功能,且两种运动方式的上述作用相似;(2)相对于持续运动,间歇运动能够明显改善SHR运动能力,其机制可能与抑制心脏重塑并无关联。Objective To compare the effects of different training modes on cardiac remodeling and exercise performance in spontaneously hypertensive rats (SHR).Methods Thirty male SHR randomly submitted to sedentary (SHR -S),interval training (SHR -IT)and continuous training (SHR -CT)groups.Ten Wistar -Kyoto (WKY)rats were used as normotensive control.After experiment,exercise performance,caudal artery pressure,cardiac structure and function were determined,and histopathological detection by HE and Masson staining was made,along with mRNA expression of heart by PCR technique. Results Compared with WKY group,rats in SHR -S group occurred elevated blood pressure,cardiac hypertrophy and collagen excessive deposition,upregulation of fetal gene expression,downregulation of contractile protein and reduced cardiac function and exercise performance (P <0.05).Compared with SHR -S group,rats in SHR -IT and SHR -CT groups showed descent blood pressure,alleviation of cardiac hypertrophy and myocardial fibrosis,downregulation of fetal gene expression, upregulation of contractile protein and enhancement of cardiac function and exercise performance (P <0.05).Compared between groups,exercise performance of SHR -IT group was higher than that of SHR -CT group (P <0.05).Conclusion (1)Both interval training and continuous training reduce blood pressure level at rest,inhibit cardiac remodeling and enhance cardiac function in SHR,whereas no major difference is observed between protocols.(2)Interval training promotes'superior improvements in exercise capacity than continuous training,which is an independence of inhibition of cardiac remodeling.
