摘  要:目的:观察补益宗气方对COPD大鼠营养状态及血清肿瘤坏死因子(TNF-α)、肌抑素(MSTN)水平的影响,探讨其改善COPD营养不良的可能作用机制。方法:将30只SD大鼠,随机分为空白组、模型组、治疗组,以气管内注射脂多糖加烟雾吸入及冷风刺激建造COPD模型大鼠,治疗组给予补益宗气方灌胃,采用酶联免疫吸附方法对各组大鼠血清前白蛋白、瘦素、TNF-α、MSTN水平进行检测。结果:(1)补益宗气方干预前,模型组、治疗组血清TNF-α及MSTN水平均明显高于空白组(P<0.05),血清前白蛋白及瘦素水平明显低于空白组(P<0.05);补益宗气方干预后,治疗组血清中TNF-α及MSTN含量比模型组显著降低(P<0.05),血清前白蛋白和瘦素水平则明显高于模型组(P<0.05);(2)补益宗气方干预后,治疗组体质量和Lee’s指数较模型组上升明显(P<0.05)。结论:补益宗气方能增加COPD大鼠体质量和Lee’s指数,改善营养状态,其作用机制可能与其降低TNF-α及MSTN水平,提高前白蛋白和瘦素水平有关。Objective: To observe the effect of Tonifying Zongqi Formula on nutritional status the level of serum TNF-αand myostatin in rats with chronic obstructive pulmonary disease(COPD) and to discuss the possible mechanism. Methods: SD rats were randomized into 3 groups: blank group, model group, treatment group. Use the smoked cigarettes, cold air stimulation plus endotracheal lipopolysaccharide injection to establish the rat model with COPD and treatment group were treated with Tonifying Zongqi Formula, and enzyme-linked immunosorbent assay(ELISA) was carried to detecte the levels of serum prealbumin, leptin, TNF-α and myostatin of all rats. Results:(1)Before the Intervention of Tonifying Zongqi Formula, the level of TNF-α and myostatin of model group and treatment group were much higher than blank group(P<0.05), while the level of serum prealbumin and leptin was significantly lower than that of control(P<0.05). After the Intervention the level of TNF-α and Myostatin of treatment group were lower than model group(P<0.05). While the level of Serum prealbumin and leptin was significantly higher than that of model group(P<0.05).(2)Compared with the model group, the body weight and Lee’s index of the treatment group after the Intervention of Tonifying Zongqi Formula increased significantly(P<0.05). Conclusion: Tonifying Zongqi Formula can increase body weight of rats with COPD and improve the nutritional status, and its mechanism of action may related to the reduction of TNF-α and the level of myostatin and the improvement of the level of prealbumin and leptin.
