摘 要:目的:观察小儿久咳合剂对感染后咳嗽(肺热阴虚证)大鼠肺匀浆中P物质(SP)含量及肺组织中SP、类香草素受体1(VR1)及蛋白基因产物9.5(PGP-9.5)表达的影响,探讨该药对大鼠气道神经源性炎症的调节作用。方法:从50只SD大鼠中随机选取10只作为空白组,其余40只采用烟熏+内毒素滴鼻+甲状腺素灌胃+辣椒素雾化诱咳法建立感染后咳嗽(肺热阴虚证)大鼠模型,造模成功后随机分为模型组、孟鲁司特钠组、小儿久咳合剂小剂量组、小儿久咳合剂大剂量组,每组10只。各组分别予以相应药物进行干预,空白组及模型组则给予同体积0.9%氯化钠溶液灌胃。ELISA法检测治疗前后各组大鼠肺匀浆中SP含量,免疫组化法检测治疗前后大鼠肺组织中SP、VR1及PGP-9.5表达情况。结果:与模型组比较,小儿久咳合剂大、小剂量组大鼠肺匀浆中SP含量明显降低(P<0.01),肺组织中SP、VR1及PGP-9.5蛋白表达也明显减少。结论:小儿久咳合剂能通过降低肺匀浆中SP含量,减少肺组织中SP、VR1及PGP-9.5的表达来减轻气道神经源性炎症,进而发挥治疗作用。Objective:To observe the effects of Pediatric Chronic Cough Mixture (PCCM) on the content of SP and the expressions of SP,VR1,PGP-9.5 in lung tissue of rats with post infectious cough (PIC) with lung yin deficiency syndrome,and to explore its regulating action on airway neurogenic inflammation.Methods:The fifty rats were enrolled and randomly divided into control group (n=10) and model group (n=40).The PIC (lung yin deficiency syndrome)model rats were made by cigarettesmoking exposure,intranasal administration with small dose of LPS,thyroid hormone and capsaicin atomization method.Then, the PIC (lung yin deficiency syndrome)model rats were randomly divided into model group (n=10),montelukast sodium group (n=10),PCCM group with low doses (n=10) and high doses (n=10).Each group was treated with the corresponding drugs,and the blank group and the model group were given the same volume of normal saline.The content of SP in lung homogenate of rats in each group before and after treatment was detected by ELISA.The expressions of SP,VR1 and PGP-9.5 in lung tissue of rats before and after treatment were detected.by immunohistochemistry.Results:Compared with the model group,the content of SP was significantly lower (P<0.01).While the expressions of SP,VR1,PGP-9.5 in rats'lung tissue in PCCM group with low dose and high dose were declined.Conclusion:PPCM can reduce airway neurogenic inflammation of rats with PIC (lung yin deficiency syndrome)by declining the content of SP and the expressions of SP,VR1,PGP-9.5 in rats'lung tissue.
