摘 要:目的研究慢性应激对阿尔茨海默病(AD)模型大鼠的影响,并探讨逍遥散对慢性应激下AD模型的干预作用。方法将大鼠随机分组,除空白组(K)外,假手术组(J)海马区注射PBS溶液,AD组在海马区注射NaN_3复制AD模型,慢性应激组(M)采用多种慢性应激刺激制备模型,而复合模型组(F)、逍遥散组(XYS)则采用慢性刺激加海马区注射NaN_3复制慢性应激下AD模型。观测行为学指标,检测血清皮质酮含量并观察各组海马组织CA1区形态学变化。结果与K组比较,M、F组自由活动受到显著影响(P<0.01),J组和AD组无统计学差异;与F组比较,逍遥散可显著改善复合模型的记忆损伤与自由活动情况(P<0.01,P<0.05)。与K组比较,F、M和AD组皮质酮含量显著升高(P<0.01);与F组比较,XYS组皮质酮含量显著降低(P<0.01)。HE染色结果显示,与K组比较,AD、M与F组的海马组织均出现了病理变化;与F组比较,逍遥散可显著改善其病理变化。结论慢性应激与NaN_3均可损伤大鼠的记忆功能,并且慢性应激能显著加重AD的记忆损伤,而逍遥散可能通过降低皮质酮含量与改善海马组织的损伤来缓解慢性应激下AD的记忆障碍。Objective To study effects of chronic stress on rats with AD; and to explore the intervention of Xiaoyaosan on AD in rats with chronic stress. Methods Rats were randomly divided into six groups. The Sham operation group (J) was injected with PBS solution in the hippocampus. The AD group was injected with NaN3 in the hippocampal area to replicate the AD model. Chronic stress model group (M) was prepared using a variety of chronic stress stimulation. In the compound model group (F) and XYS group,chronic stimulation plus injection of NaN3 in the hippocampus region was used to replicate the AD model with chronic stress. The behavioral changes of rats were observed, serum corticosterone was detected, and morphological changes in CA1 area of hippocampal tissue were observed. Results Compared with the control group (K),chronic stress significantly affected the free activity of group M and F (P<0.01). There was no statistically significant difference between group K and group J or AD. Compared with group F, Xiaoyaosan significantly improved the memory damage and free activity of the compound model (P<0.01,P<0.05). Compared with group K,the contents of corticosterone in F,M and AD groups increased significantly (P<0.01). Compared with group F,the content of corticosterone in XYS group was significantly decreased (P<0.01). Compared with group K, the hippocampal tissues of groups AD, M and F showed pathological changes. Compared with group F,Xiaoyaosan significantly improved the pathological changes. Conclusion Both chronic stress and NaN3 can damage the memory function of rats; and chronic stress cansignificantly aggravate the memory impairment of AD. Xiaoyaosan may reduce corticosterone content and improve the hippocampus damage,to improve memory disorders of AD rats with chronic stress.
