摘 要:目的研究新疆栽培荒漠肉苁蓉水提物(aqueous extracts of cultivated Cistanche deserticola Y.C.Ma in Xinjiang,AECCD)对小鼠T细胞应答和抗体持久性的影响,探讨其对小鼠的免疫增强作用。方法选取ICR雌性小鼠60只(共两批小鼠,每批30只),每批随机分为6组,每组5只,分别将低、中、高3个剂量的AECCD配伍模式抗原卵清白蛋白(ovalbumin,OVA)为实验组,0.9%NaCl为空白对照组,OVA单独免疫组为阴性对照组,铝佐剂(aluminium adjuvant,Alum)作为阳性对照组,肌肉免疫2次,间隔2周。采用流式细胞术检测T细胞亚群、细胞因子及树突状细胞(dendritic cells,DC)表面分子的表达水平;间接ELISA法检测小鼠血清中的IgG抗体水平。结果AECCD能够显著增强脾细胞中CD4+、CD8+T细胞亚群的比例(P<0.05),显著提高CD4+CD44+和CD8+CD44+效应性T细胞的表达水平(P<0.05);促进T细胞IFN-γ的分泌,也能够促进DC表面分子CD40和CD80的表达水平(P<0.05);ELISA检测结果表明高剂量AECCD可以显著提高OVA诱导的IgG抗体表达水平的持久性(P<0.05)。结论AECCD能增强OVA蛋白诱导的T细胞免疫应答,保持抗体的持久性,从而提高机体的免疫应答水平。Objective To study the effects of aqueous extracts of cultivated Cistanche deserticola Y. C.Ma (AECCD)on T cell responses and the duration of antibody response and to investigate its immunoen- hancing activities in mice.Methods Two batches of female ICR mice were used in this study with 30from each batch.Each batch of mice was randomly divided into six groups (n=5).Low,medium and high doses of AECCD in combination with ovalbumin (OVA)were used to set up three experimental groups,while 0.9% NaCl,OVA alone and aluminium adjuvant were respectively used as blank,negative and positive controls.All mice were intramuscularly injected twice at an interval of two weeks.Flow cytometry was used to detect the expression of T lymphocyte subsets,cytokines and surface molecules of dendritic cells (DC).Indirect ELISA was used to detect IgG antibody levels.Results AECCD could significantly increase the percentage of CD4+and CD8+T lymphocytes in spleen (P<0.05),up-regulate the expression of CD4+CD44+and CD8+CD44+effector T lymphocytes (P<0.05),promote the secretion of IFN-γ in T lymphocytes and enhance the expression of CD40 and CD80on the surface of DC (P<0.05).ELISA results showed that high-dose AECCD could significantly prolong the duration of IgG antibody response induced by OVA (P<0.05).Conclusion AECCD could en- hance the T lymphocyte immune response induced by OVA and keep it maintained at a high level ,.which might help to improve the body's immune response.
