摘  要:目的建立同时测定玉叶解毒颗粒中绿原酸和槲皮素两种主要成分含量的HPLC方法。方法采用Phenomenonex Gemini C18色谱柱(250 mm×4. 6 mm,5μm),以甲醇(A)-0. 4%磷酸水溶液(B)为流动相,梯度洗脱(0~20 min,80%A→55%A; 30~45 min,15%A→0%A),流速1 m L/min,检测波长为345 nm。结果玉叶解毒颗粒中绿原酸、槲皮素的含量范围分别为:0. 30~3. 00μg/g,0. 18~1. 8μg/g,且线性关系良好;平均回收率分别为99. 930%、95. 005%,RSD均小于2. 0%。结论该检测方法简便,可靠,稳定。经方法学验证,可用于同时检测玉叶解毒颗粒中绿原酸和槲皮素的含量。Objective To establish an HPLC method for simutaneous determination of chlorogenic and quercitrin in Yuye Gran- ules.Methods HPLC experiments were performed on a Phenonmenonex Gemini C18 column (250mm×4.6mm,5μm),the mobile phase consisted of methanol (A)-0.4%phosphoric acid (B)in gradient mode (0-20min,80%A -55% A;30-45 min,15%A-0%A),the flow rate was 1mL/min.The detection wavelength was set at 345nm.Results The contents of chlorogenic acid and quericitrin in Yuye Granules were 0.30-3.00μg/g and 0.18-1.8μg/g,showed a good linear relation- ship.The average recoveries of chlorogenic and quercitrin were 99.930%and 95.005%with RSD below 2.0%. Conclusion The detection method was simple,reliable,stable.The establish method can be used in dertermination of chlorogenic acid and quercitrin in Yuye Granules.
