摘 要:目的采用响应面法优化蟾酥固体脂质纳米粒(CS-SLN)制备工艺。方法采用乳化蒸发-低温固化法制备CSSLN,以包封率、载药量、粒径及综合值为指标,考察载体(GME)用量、大豆磷脂用量及泊洛沙姆188(P188)用量对制备工艺的影响。同时运用Box-Behnken响应面法优化制剂处方,并对结果进行方程拟合,预测最佳工艺条件。结果制备60m L蟾酥固体脂质纳米粒混悬液的最优处方构成为单硬脂酸甘油酯0. 56 g,大豆磷脂量2. 24 g,泊洛沙姆188 0. 5 g,根据优化方案制备的蟾酥固体脂质纳米粒包封率为86. 4%,载药量为1. 88%,平均粒径为116. 3 nm。结论该处方可用于蟾酥固体脂质纳米粒的制备,工艺简单,Box-Behnken响应面法可用于蟾酥固体脂质纳米粒的工艺优化,建立的模型具有良好预测性。Objective To optimize the preparation process of the toad venom-loaded solid lipid nanoparticles(CS-SLN) by response surface method. Methods The CS-SLN was prepared by the method of emulsion-evaporation and solidification. And take the encapsulation rate,drug-polymer interactions,particle size and composite value as an indicator,to examine the effect of the dosage of GME,soybean phospholipid and Poloxamer 188(P188) on preparation technology. At the same time,the Box Behnken response surface method was used to optimize the prescription,and the results were fitted into equation,to predict the best process conditions. Results The preparation of 60 m L toad venom solid lipid nanoparticles(CS-SLNS) was detected the optimal prescription need GME 0. 56 g,soybean phospholipid 2. 24 g,and P188 0. 5 g. According to the optimized scheme,the encapsulation efficiency and drug loading rate of CS-SLN is 86. 4% and 1. 88%,and the average particle size was 116. 3 nm. Conclusion This prescription can be used for the preparation of CS-SLN,the Box-Behnken response surface method can be used to optimize the process of the solid lipid nanoparticles,and the model has good predication.
