摘  要:目的对对叶百部Stemona tuberosa Lour.不同居群进行遗传多样性和遗传结构进行分析。方法利用荧光AFLP标记技术,8对EcoR I/Mse I引物组合对38份供试材料的基因DNA扩增,进行多态性检测,用NTSYSpc-2. 11F软件进行数据分析。以DICE相似系数为基础而得出UPGMA个体间聚类图和相似系数。结果共获得条带数1477条,多态性条带数1477条,多态性达到了100%。UPGMA个体间聚类结果表明,对叶百部种群间的遗传距离与地理距离有相关性。结论对叶百部不同居群间的遗传多样性非常丰富。为对叶百部种质资源的合理利用和品种选育提供了分子水平的理论依据。Objective To analyze the genetic diversity and genetic structure of different population of Stemona tuberosa Lour.Methods 8 primer combinations(EcoR I/Mse I) were used to analyze the Amplified Fragment Length Polymorphism(AFLP) in38 test materials. Polymorphisms were detected and the data was analyzed by NTSYSpc-2. 11 F software,then UPGMA clustering figure and similarity coefficient based on DICE were given. Results A total of 1477 polymorphic bands were dectected and the polymorphism was 100%. The clustering results showed that there was a correlation between the genetic distance and geographical distance of Stemona tuberosa Lour. populations. Conclusion There were high genetic diversity and rich polymorphism in Stemona tuberosa Lour. population. The study provided a theoretical basis at the molecular level for the rational use of germplasm resources of Stemona tuberosa Lour. and their new variety breeding.
