摘  要:目的探索乙酰半胱氨酸对骨髓成纤维细胞转化生长因子-β( transforming growth factor-β,TGF-β)和I型胶原的抑制作用。方法选择健康成年远交群(spraguedawley.SD)大鼠雌雄不限,将36只大鼠随机分成正常血清组和乙酰半胱氨酸血清组,每组18只,经灌胃和DMEM培养基稀释分别制 备正常血清和乙酰半胱氨酸血清;以大鼠骨髓组织体外培养成纤维细胞为材料,经免疫荧光鉴定后采用 MTT法检测成纤维细胞增殖率,酶联免疫吸附法试验(enzyme linked immunosorbent assay,EUSA)和实时 荧光定量PCR法检测TGF-β;和I型胶原表达水平。结果乙酰半胱氨酸血清组成纤维细胞增殖率显 著低于正常血清组[(42.51±2.62)% 比(71.62±4. 51)%=21. 635,P<0.05],TGF-β和I型胶原表 达水平显著低于正常血清组[(27.91± 5.73 ) μmol/L 比(35.61±6. 42)μmol/L,t= 10.235P<0.05;(21.21±4.62)μmol/L 比(28.72±5.91)μmol/L,t = 9. 658,P<0. 05] ,mRNA 表达水平显著低于正常血 清组[(0.25±0.10)比(0.46±0.11)= 12.525,P<0.05; (0.13 ±0.06)比(0.40±0.10),t=8.652,P<0.05]。结论乙酰半胱氨酸对骨髓成纤维细胞TGF-β和I型胶原有一定抑制作用。Objective To explore the inhibitory effect of N-acetylcysteine ( NAC) on transforming growth factor β(TGF-β) and type I collagen in bone marrow fibroblasts. Methods the male and female Sprague Dawley (SD) rats were selected and randomly divided into normal serum and N-acetylcysteine( NAC) serum group with 18 SD rats for each group. Both were prepared by gastrostomy and diluted by DMEM medium, and the fibroblasts were cultured in vitro. The proliferation rate of fibroblasts was detected by MTr method after identified by immunofluorescence. The expressions of TGF-p and type I collagen were detected by enzyme linked immunosorbent assay (ELISA) and quantitative real-time PCR(qPCR). Results The proliferation rate of fibro-blasts from N-acetylcysteine( NAC) serum was significantly lower than that of normal serum group [ (42. 51 ± 2. 62)% vs. (71.62 ±4. 51 )%,t=21.635,P <0. 05],TGF-β and type I collagen expression level was sig-nificantly lower than that of normal serum group [ (27.91 ±5.73) ratio (35.61 ±6.4)2 μmol/L,f = 10. 235, P<0. 05; (21.21 ±4. 62) vs. (28. 72 ±5. 91)=9. 658,P <0. 05 ] ,mRNA expression level was significant-ly lower than that of normal serum group [(0.25 ±0. 10) vs. (0.46 ±0. 11 )t= 12. 525,P <0. 05 ;(0.13 ± 0? 06) vs. (0.40 ±0. 10) t=8. 652,P <0. 05 ]. Conclusion N-acetylcysteine( NAC) could have certain inhibitory effects on the proliferation of bone marrow fibroblasts TGF-β and type I collagen.
