摘 要:目的:建立同时测定薄荷药材中5种有机酸和黄酮类成分含量的高相液相色谱方法,比较不同产地薄荷药材各成分的差异。方法:采用HPLC-PDA法,Waters XBridge C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%甲酸水溶液,流速为1.0 m L/min,柱温30℃,检测波长285 nm,进样量10μL。采用灰色关联度分析(GRA)对其进行综合评价。结果:5种有机酸和黄酮类成分在一定范围内线性关系良好,相关系数均大于0.9999;平均加样回收率95.76%~103.00%,相对标准偏差为0.8%~2.41%。同一产地中的不同成分、不同产地中的同一成分均存在一定差异,安徽-太和-1的薄荷样品综合品质评价最优。结论:该实验建立的方法准确可靠,可同时测定薄荷样品中咖啡酸、迷迭香酸、橙皮苷、蒙花苷、香叶木素的含量,为薄荷非挥发性成分的测定提供了可行方案。Objective: To establish an HPLC method for simultaneous determination of five kinds of organic acids and flavonoids inMentha haplocalyx from different regions. Methods: The analysis was carried on a Waters XBridge C18 column(250mm×4.6mm,5μm)with elution by mobile phase of methanol-0. 1% formic acid in water at a flow rate of 1.0 mL/min. The column temperature was maintainedat 30℃. The detection wavelength was set at 285 nm while the injection volume was 10 μL. Grey relational analysis(GRA)wasused to comprehensively evaluate the quality of the samples. Results: The five components showed good linear relationships in the linearranges,the correlation coefficient was greater than 0. 9999. The average recoveries of the five components were 95.76%~103.00% andthe relative standard deviations were 0.8%~2.41%. There were certain differences in different components of the same region and thesame component in different regions. The comprehensive evaluation of Mentha haplocalyx from Anhui-Taihe-1 was the best. Conclusion:The method established in this experiment is able to simultaneously determine the content of caffeic acid,rosmarinic acid,hesperidin,anthocyanin and geraniol in samples of Mentha haplocalyx.It provides a feasible scheme for the determination of the nonvolatile componentsof Mentha haplocalyx.
