摘 要:目的:优选混合酶法提取五指毛桃多糖的工艺。方法:通过单因素试验设计,以多糖收率为指标,考察不同影响因素对工艺的影响,并根据单因素结果通过D-最优混料设计优化复合酶的配比,用Box-Behnken试验设计法对得出的影响因素进行筛选。结果:混合酶最佳比例为果胶酶占25.2%、纤维素酶占66.7%、甘露聚糖酶占8.1%;混合酶法提取五指毛桃多糖最优工艺为加酶量5.0%、溶媒倍数36倍、反应体系p H 4.7、酶反应温度42.5℃及酶反应时间294 min,五指毛桃多糖收率在1.39%左右。结论:利用Box-Behnken效应面法优选的五指毛桃多糖复合酶提取工艺稳定可行,可用于五指毛桃多糖的提取。Objective:To optimize conditions of enzymatic extraction of Ficus simplicissima.Methods:Through the single factor experiment design,the effect of different factors on the process was investigated with the yield of polysaccharide as the index,optimization of compound enzyme ratio by D-optimal mixture design based on single factor results.Based on the response surface methodology by using Box-Behnken design,different influence factors were examined.Results:The optimal combination of enzymes were 25.2% pectinase,66.7% cellulase and 8.1% mannose.The dosage of the mixed enzyme was 5.0%,the solvent ratio was 36,initial reaction system pH was 4.7,enzymatic hydrolysis temperature was 42.5℃,and enzymatic hydrolysis time was 294 min.Under those conditions,the yield of polysaccharides of Ficus simplicissima was 1.39%.Conclusion:The Box-Behnken response surface methodology which used in the experiment to optimize the enzymatic extraction process for the polysaccharide of Ficus simplicissima is stable,effective and feasible.
