摘 要:目的:研究藏药波棱瓜子总木脂素对大鼠慢性酒精性肝损伤的保护作用及其机制。方法:60只雄性Wistar大鼠按体质量随机分成正常对照组、模型组、易善复(95 mg/kg)阳性对照组及波棱瓜子总木脂素高(400 mg/m L)、中(200 mg/m L)、低(100 mg/m L)剂量组。各给药组每日上午9点给予2 m L相应药物,0.5 h后,除正常对照组外各组给予56度白酒灌胃,连续造模8 w后,测定各组大鼠血清ALT、AST及肝脏GSH-Px、MDA、SOD、GSH、CAT、TG、ADH、ALDH2水平。荧光定量PCR法测定大鼠肝微粒体CYP2E1 mRNA表达,蛋白质印迹法(Western blot)测定肝微粒体CYP2E1蛋白表达。观察肝组织病理学改变。结果:与模型组比较,除低剂量组TG及高剂量组GSH-Px差异无统计学意义(P>0.05)外,波棱瓜子总木脂素各组大鼠血清ALT、AST水平显著降低;肝组织MDA、TG、ADH水平及肝微粒体CYP2E1 mRNA和蛋白表达显著降低;肝组织SOD、GSH、GSH-Px、CAT、ALDH2水平显著升高(P<0.05或P<0.01)。HE显示与模型组比较波棱瓜子总木脂素各组肝组织破坏程度较轻。结论:藏药波棱瓜子总木脂素对大鼠慢性酒精性肝损伤有保护作用。Objective:To study the hepatoprotective effect and mechanism of the total lignans from tibetan medicinal Herpetospermum pedunculosum seeds against alcohol-induced chronic liver injury in rats.Methods:60 male Wistar rats were randomly divided into six groups ,normal control group,model group ,positive control group(95 mg/kg)and Herpetospermum pedunculosum seeds total lignans groups(400,200,100 mg/kg).The administration groups were given the designed medicine(2 mL)at 9:00 am.At 0.5 h post administration,except for the control group,the other five groups were given 56°liquor for 8 w.The levels of serum alanine aminotransferase(ALT),serum aspartate aminotransferase(AST),and the liver levels of glutathione peroxidase(GSH-Px),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione(GSH),catalase(CAT),triglycerides(TG),ADH and ALDH2 were measured.The expression of CYP2E1 mRNA in rat liver microsomes were measured by the real-time fluorescence quantitative PCR.The expression of CYP2E1 protein in liver microsomes were detected by Western blot.The pathological changes of liver tissue were also observed.Results :Compared with the model group,in addition to the low dose group TG and high dose group GSH-Px,there was no stasistical significance(P>0.05),the serum ALT and AST activities of the total lignans groups decreased significantly.The liver levels of MDA,TG,ADH, CYP2E1 mRNA and protein expression levels decreased in total lignans groups.The SOD,GSH,GSH-Px,CAT and ALDH2 levels increased significantly in total lignans groups(P<0.05or 0.01).According to HE staining,each total lignans groups showed different degrees of hepatic damage.Partial necrosis of liver cells,but the damage was lighter than that of the model group.Conclusion:The tibetan medicinal Herpetospermum pedunculosum's total lignans can protect liver from the chronic alcoholic damage.
