摘 要:目的:比较吴茱萸不同炮制品的肝脏毒性作用,探讨吴茱萸的炮制减毒机制。方法:将小鼠分为正常组和吴茱萸不同炮制品组(生吴茱萸、甘草制吴茱萸及盐制吴茱萸),各炮制品又分为6.5、3.25、0.65 g/kg剂量组,各组连续ig 15 d后处理,检测小鼠血清ALT、AST活力,肝组织中SOD、MDA、Na^+,K^+-ATP酶、Ca^(2+)-ATP酶、TNF-α、IL-1β水平,免疫组化法观察肝组织中Bcl-2、Bax蛋白表达。结果:与正常对照组比较,生吴茱萸组能升高血清中ALT、AST含量,降低肝组织Na^+,K^+-ATP、Ca^(2+)-ATP、SOD活力,升高MDA、TNF-α、IL-1β水平,肝组织Bcl-2蛋白表达减弱,Bax蛋白表达增强;与生吴茱萸组比较,甘草制吴茱萸组、盐制吴茱萸组能降低血清ALT、AST含量,升高肝组织Na^+,K^+-ATP、Ca^(2+)-ATP活力、SOD活力,降低MDA、TNF-α、IL-1β水平,肝组织Bcl-2蛋白表达增强,Bax蛋白表达减弱。结论:甘草制和盐制可有效降低吴茱萸的肝毒性,减轻吴茱萸导致的肝损伤,其炮制减毒的机制可能与减轻肝组织氧化损伤,改善肝组织能量代谢,减少炎症因子释放以及调节细胞凋亡相关蛋白Bcl-2和Bax的表达有关。Objective:To assess the hepatotoxicity of Euodia rutaecarpa prepared with different methods and to investigate the mechanisms of attenuation by different preparing ways.Methods :The mice were divided into normal group and treated group which were received different prepared Euodia rutaecarpa fruit(raw products,licorice products and salt products).The prepared products were divided into three different dose groups and were given in dose of 6.5,3.25and 0.65g/kg for 15 days ,respectively.The activity of ALT and AST in serum and the levels of SOD,MDA,Na^+,K^+-ATPase,Ca^2+-ATPase,TNF-α and IL-1β in liver tissue were assayed.The ex- pression of Bcl-2 and Bax protein in liver tissue was observed by immunohistochemical method.Results:Compared with the normal group,the levels of ALT and AST in serum increased and the activities of Na^+,K^+-ATPase,Ca^2+-ATPase as well as SOD in liver tissue decreased.Besides,the levels of MDA ,TNF-α and IL-1β increased and the expression of Bcl-2 protein decreased.Additionally,the ex-pression of Bax protein was enhanced in raw product group.Compared with the raw product group,the contents of ALT and AST in ser- um decreased and the activities of Na^+,K^+-ATPase,Ca^2+-ATPase and SOD in liver tissue were increased.Moreover,the levels of MDA, TNF-α and IL-1β were decreased.The expression of Bcl-2 protein was enhanced and the expression of Bax protein was decreased in the licorice group as well as in the salt product group.Conclusion :After prepared by licorice and salt,the hepatotoxicity of Euodia rutaecarpa fruit is reduced dramatically and the liver damage caused by Euodia rutaecarpa can be curbed.The mechanisms of attenuation of Euodia rutaecarpa by different prepared methods may be related to alleviation of the oxidative damage to liver tissue and the products can improve the energy metabolism of liver tissue.Furthermore,they can reduce the release of inflammatory factor and regulate the expres- sion of apoptosis protein Bcl-2 and Bax.
