摘 要:目的:探讨芳香新塔花总黄酮(ZCF)对过氧化氢(H2O2)诱导人脐静脉内皮细胞(HUVECs)损伤的保护作用。方法:采用体外培养HUVECs,分为正常组、模型组、芳香新塔花总黄酮(25、50、100μg/ml)组和阳性药维生素C组。采用CCK-8法检测HUVECs活力,用试剂盒检测各组细胞丙二醛(MDA)、乳酸脱氢酶(LDH)和还原型谷胱甘肽(GSH)含量、超氧化物歧化酶(SOD)的活性。采用流式细胞术检测活性氧(ROS)和Hoechst33258荧光染色观察细胞凋亡情况。结果:与正常组比较,模型组细胞存活率、GSH含量和SOD活性明显降低,MDA、LDH和ROS含量明显增加,细胞凋亡率显著上升;与模型组比较,芳香新塔花总黄酮(25、50、100μg/ml)组和维生素C组可显著提高细胞存活率和SOD活性,明显增加GSH含量、减少MDA和LDH含量,抑制ROS的生成,显著降低细胞凋亡率。结论:芳香新塔花总黄酮对H2O2诱导的HUVECs损伤具有保护作用,机制可能与其增强抗氧化能力、降低ROS产生和抑制细胞凋亡有关。Objective:To investigate the protective effect of Ziziphora clinopodioides.flavonoids(ZCF) on the human umbilical vein endothelial cells(HUVECs) injured by hydrogen peroxide(H2O2).Methods:HUVECs cells were cultured in vitro and were divided into six groups as follows:the normal group,the model group,Ziziphora clinopodioides flavonoids groups(25,50,100μg/ml),and positive-drug vitamin C group.CCK-8 was used to determine the HUVECs viability.The levels of Malonic dialdehyde(MDA),lactate dehydrogenase(LDH) and glutathione(GSH),and the activity of superoxide dismutase(SOD) were detected by spectrophotometric methods in the assay kit.The reactive oxygen species(ROS) of HUVECs were analyzed by flow cytometry.Apoptosis was observed by Hoechst 33258 fluorescent staining.Results:Compared with the normal group,the model group significantly decreased HUVECs viability,the level of GSH and the activity of SOD(In the model group,HUVECs viability,the level of GSH and the activity of SOD significantly decreased),the levels of MDA,LDH and ROS significantly increased,the apoptosis rate significantly increased(P<0.01).Compared with the model group,Ziziphora clinopodioides.flavonoids(25,50,100μg/ml) and vitamin C significantly increased HUVECs viability and the activity of SOD(P<0.05,P<0.01) in a dosedependent manner,it significantly increased the levels of GSH,significantly decreased the levels of MDA and LDH,and inhibited ROS production(P<0.05,P<0.01),significantly decreased the apoptosis rate(P<0.01).Conclusion:Ziziphora clinopodioides.flavonoids has the significant protective effect on HUVECs injured by H2O2,and the underlying mechanism may be related to enhancing the antioxidant capacity,reducing ROS production and apoptosis inhibition.
